摘  要:为了了解四川地区流行的猪流行性腹泻病毒(PEDV)的生物学特征及基因的遗传进化关系,从四川某地采集了发病仔猪小肠,病料处理后在Vero细胞上进行盲传,通过RTPCR、间接免疫荧光鉴定(IFA)验证,成功分离出1株猪流行性腹泻病毒,命名为X-6/2021株。对X-6/2021株S1基因进行RT-PCR扩增,获得的分离株S1序列与国内外PEDV毒株进行序列比对和遗传进化分析。结果显示,PEDV-X-6/2021毒株与Ⅰ型参考毒株的同源性为90.6%~93.2%,与Ⅱ型参考毒株的同源性为97.6%~99.0%。试验成功地从四川省分离鉴定得到1株猪流行性腹泻病毒,为该地区PEDV防控及候选疫苗毒株的筛选提供了物质基础。In order to further study the characteristics of the current porcine epidemic diarrhea virus(PEDV)and its genetic evolutionary relationship in Sichuan Province,the feces and small intestine contents of piglets were collected from Sichuan Province,inoculated and cultured in the Vero cells.By RT-PCR and indirect immunofluorescence assay(IFA),a PEDV isolate was successfully identified and named as PEDV-X-6/2021 strain.The S1 gene of X-6/2021 strain was amplified by RT-PCR and analyzed along with the sequence alignment and phylogenetic trees of PEDV S1 gene sequences from other domestic and international strains.The results showed that the S1 gene of X-6/2021 strain had 90.6%-93.2%nucleotide homology with typeⅠstrains,and 97.6%-99.0%nucleotide homology with typeⅡstrains.The results showed that a strain of porcine epidemic diarrhea virus in Sichuan Province was successfully isolated and identified in this experiment,which provided a material basis for the selection of tporcine epidemic diarrhea vaccine strains in this area.
