超声心动图评价小鼠主动脉弓缩窄模型心脏结构与功能

摘  要:目的建立因小鼠主动脉弓缩窄所致的心衰模型,应用高频超声波心动图及实验室技术评价该模型的成功性及其改变特点。方法本次实验选取C57BL/6J的小鼠20只,随机分为手术组(transverse aortic constriction,TAC)、假手术组(sham-operation,SHAM)。手术组施行主动脉弓缩窄术建立压力超负荷心衰模型。分别于术前1 d、术后1 w、术后6 w应用高频超声心动图测量小鼠左心室结构和收缩功能参数;术后6 w时进行心脏及体重称重,记算出心体比值;蛋白印迹检测心肌肥大标志物,心房利钠肽(atrial natriuretic peptide,ANP)蛋白表达水平;通过马松(Masson)、苏木素-伊红(HE)染色,对两组小鼠的心肌病理组织学变化进行观测。结果与对照组进行比较,实验6 w时超声心动图的数据显示,手术组小鼠左心室大小、心腔容积均有所增加(P<0.05),左心室射血分数和短轴缩短率降低较为明显(P<0.001);心脏重量、心体比值(HW/BW)较对照组均升高(P<0.05);HE及Masson染色见心肌细胞肥大、胶原纤维含量增加,ANP蛋白表达水平升高。结论高频超声心动图能够显示小鼠压力诱导性心衰过程中心脏结构与功能的特征性改变,为以后小鼠主动脉弓缩窄心衰模型的研究操作提供理论依据。Objective To establish the mouse model of heart failure induced by transverse aortic constriction and evaluate the success and the change characteristics of the model by high-frequency echocardiography and laboratory techniques.Methods In this experiment,20 C57BL/6J mice were selected and randomly divided into operation group and sham operation group.The operation group underwent transverse aortic constriction to establish a pressure overload heart failure model.The parameters of left ventricular structure and systolic function were measured by high-frequency echocardiography at 1 day before operation,1 week and 6 weeks after operation.At 6 weeks after operation,the heart and body weight were weighed,and the heart-body ratio was recorded.The protein expression of atrial natriuretic peptide(atrial natriuretic peptide,ANP),a marker of myocardial hypertrophy,was detected by Western blot.Masson and hematoxylin-eosin(HE)staining were used to observe the myocardial histopathological changes of the two groups of mice.Results Compared with the control group,the echocardiographic data at 6 weeks showed that the left ventricular size and cardiac chamber volume in the operation group were increased(P<0.05),and the left ventricular ejection fraction and fractional shortening were decreased significantly(P<0.001).The heart weight and heart body ratio in the control group were higher than those in the control group(P<0.05).HE and Masson staining showed cardiomyocyte hypertrophy,increased collagen fiber content,and increased ANP protein expression.Conclusion High-frequency echocardiography can show the characteristic changes of cardiac structure and function in the process of stress-induced heart failure in mice,which provides a theoretical basis for the future research and operation of mouse heart failure model with aortic arch constriction.
