摘  要:多糖是药用植物铁皮石斛(Dendrobium officinale)的主要成分之一。鼠李半乳糖醛酸聚糖Ⅰ鼠李糖转移酶(rhamnogalacturonⅠrhamnosyltransferase,RRT)在Ⅰ型鼠李半乳糖醛酸聚糖的合成中起重要作用。利用生物信息学在全基因组水平鉴定出4个铁皮石斛RRT家族基因(DoRRT1、DoRRT2、DoRRT3、DoRRT4),通过逆转录定量PCR检测该基因家族在不同组织和胁迫下的表达情况。结果表明:在系统发育上DoRRT1、DoRRT3被聚为一组,DoRRT2、DoRRT4被聚为一组;DoRRT蛋白均为亲水蛋白质,可定位于高尔基体;DoRRT蛋白的α-螺旋和无规卷曲较多;4个DoRRT蛋白均有O-FucT保守结构域;组织表达显示DoRRT1、DoRRT2、DoRRT4在根、茎、叶中均有表达,且在地上部的转录水平较高;胁迫表达表明DoRRT2、DoRRT4在高温胁迫下表达显著下调,DoRRT2在PEG模拟干旱条件下表达显著下调,说明DoRRT2、DoRRT4参与非生物胁迫响应。以上结果为深入研究DoRRT的生物学功能提供数据支撑。Polysaccharide is one of the main components of the medicinal plant Dendrobium officinale.rhamnogalacturonⅠrhamnosyltransferases(RRTs)are required in the synthesis of rhamngalacturonanⅠ.In this study,four D.officinale RRTs(DoRRT1,DoRRT2,DoRRT3,DoRRT4,)were identified at the genome-wide level and subjected to bioinformatics analysis.The reverse transcription quantitative PCR was used to detect the expression of DoRRTs in different tissues or under different stresses.The results showed that DoRRT1 and DoRRT3 were clustered into one group and DoRRT2,DoRRT4 were clustered into another group in phylogeny.DoRRTs were hydrophilic proteins and might be located in the Golgi apparatus.Alpha helices and random coils are the main structure of DoRRTs.O-FucT conserved domains were found in four DoRRTs.Tissue expression analysis showed that DoRRT1,DoRRT2 and DoRRT4 were expressed in roots,stems and leaves,and the transcription level in the shoots was highest.Stress expression analysis showed that the expression of DoRRT2 and DoRRT4 was significantly down-regulated under high temperature stress,while the expression of DoRRT2 was significantly down-regulated under PEG-simulated drought condition,indicating that DoRRT2 and DoRRT4 are involved in abiotic stress response.The results provide data for in-depth study of the biological functions of DoRRTs.
