摘  要:本试验旨在研究桑叶提取物对肉鸡腹脂沉积及其相关代谢指标的影响。选用192羽1日龄健康、体型相近的爱拔益加(AA)肉鸡,随机分为4组,每组6个重复,每个重复8羽。对照组饲喂基础饲粮,试验组分别饲喂在基础饲粮中添加400、800和1 200 mg/kg桑叶提取物的试验饲粮。试验期56 d,分2个阶段,即1~21日龄(前期)和22~56日龄(后期)2个阶段。本试验检测了肉鸡生长性能、血清和肝脏中与脂肪代谢相关的指标。结果表明:与对照组相比,1)饲粮中添加不同水平的桑叶提取物对肉鸡全期的平均日采食量(ADFI)、平均日增重(ADG)和料重比(F/G)均无显著影响(P>0.05);2)饲粮中添加不同水平桑叶提取物均显著降低肉鸡2个阶段的腹脂沉积量(P<0.05);3)饲粮中添加800和1 200 mg/kg桑叶提取物分别显著降低了前期和后期血清中总胆固醇(TC)含量;显著降低肝脏中TC含量(P<0.05);4)饲粮中添加不同水平桑叶提取物均显著提高了肝脏中腺苷酸活化蛋白激酶α(AMPKα)的mRNA相对表达量(P<0.05),饲粮中添加800和1 200 mg/kg的桑叶提取物显著降低了肝脏中调控脂肪酸合成的固醇调节元件结合蛋白-1c(SREBP-1c)、乙酰辅酶A羧化酶(ACC)和脂肪酸合成酶(FAS)的mRNA相对表达量(P<0.05)。综上所述,饲粮中添加400、800和1 200 mg/kg的桑叶提取物可以降低AA肉鸡腹脂沉积,其发挥降脂功效的作用原理是:显著上调肝脏中AMPKα基因表达和下调SREBP-1c、FAS、ACC等脂肪代谢相关基因的表达。通过减少脂肪的合成,减少肝脏和血清中TC的含量。因此,其作用机理与AMPK/SREBP-1c/ACC通路有关。This experiment was conducted to investigate the effects of mulberry leaf extract on abdominal fat deposition and related metabolic indexes of broilers.A total of 192 one-day-old healthy AA broilers with similar body size were randomly divided into 4 groups with 6 replicates per group and 8 broilers per replicate.Broilers in control group were fed a basal diet,and those in experimental groups were fed the basal diet supplemented with 400,800 and 1 200 mg/kg mulberry leaf extract,respectively.The experiment lasted for 56 days and was divided into two stages:1 to 21 days of age(early stage)and 22 to 56 days of age(late stage).The growth performance,serum and liver lipid metabolism related indexes of broilers were detected.The results showed as follows:compared with control group,1)dietary mulberry leaf extract had no significant effects on average daily feed intake(ADFI),average daily gain(ADG)and feed/gain(F/G)of broilers during the whole period(P<0.05);2)adding mulberry leaf extract significantly decreased the abdominal fat deposition of broilers in two experimental stages(P<0.05);3)adding 800 and 1 200 mg/kg mulberry leaf extract significantly reduced serum total cholesterol(TC)content in early and late stage,respectively;TC content in liver was significantly decreased(P<0.05);4)adding mulberry leaf extract significantly increased the mRNA relative expression level of AMP-activated protein kinaseα(AMPKα)in liver(P<0.05),adding 800 and 1 200 mg/kg mulberry leaf extract significantly decreased the mRNA relative expression levels of sterol regulatory element binding protein-1c(SREBP-1c),acetyl-CoA carboxylase(ACC)and fatty acid synthetase(FAS)in liver(P<0.05).In conclusion,adding 400,800 and 1 200 mg/kg mulberry leaf extract can reduce abdominal fat deposition of AA broilers.The mechanism of lipid lowering effect of mulberry leaf extract is as follows:mulberry leaf extract significantly increases the expression of AMPKα gene in liver,and significantly lower the expression of SREBP-1c,FAS,ACC genes in liver which
