摘 要:目的探究化浊解毒方治疗慢性肾功能衰竭(chronic renal failure,CRF)的机制。方法将60只SD大鼠随机分为正常组、模型组、尿毒清组、氯沙坦钾组、化浊解毒组,采用5/6肾切除术复制CRF模型。模型复制后连续干预12周,苏木精-伊红染色后光学显微镜下观察肾脏组织形态学变化,分别采用Western blot法、RT-PCR法检测肾组织转化生长因子β1(transforming growth factor beta 1,TGF-β1)、α-平滑肌肌动蛋白(alpha-smooth muscle actin,α-SMA)、肝细胞生长因子(hepatocyte growth factor,HGF)蛋白及其mRNA的表达水平。结果与对照组比较,模型组大鼠血清肌酐(serum creatinine,SCr)、血尿素氮(blood urea nitrogen,BUN)和肾组织TGF-β1、HGF、α-SMA蛋白及其mRNA表达水平明显升高(P<0.05);与模型组比较,尿毒清组、氯沙坦钾组、化浊解毒组大鼠SCr、BUN及肾组织TGF-β1、α-SMA蛋白及其mRNA表达水平明显降低(P<0.05),肾组织HGF蛋白及其mRNA表达水平明显升高(P<0.05)。尿毒清组、氯沙坦钾组、化浊解毒组大鼠肾脏纤维化水平有不同程度改善。结论化浊解毒方可有效改善CRF大鼠肾功能,其机制可能与抑制CRF大鼠肾脏组织中TGF-β1的表达并上调HGF的表达,从而抑制α-SMA的表达有关。Objective To investigate the mechanism of action of Huazhuo Jiedu Prescription in the treatment of chronic renal failure(CRF).Methods A total of 60 Sprague-Dawley rats were randomly divided into normal group,model group,Uremic Clearance Granule group,losartan group,and Huazhuo Jiedu group,and 5/6 nephrectomy was used to establish a model of CRF.After intervention for 12 consecutive weeks,HE staining was used to observe renal histomorphological changes under a light microscope,and Western blot and RT-PCR were used to measure the protein and mRNA expression levels of transforming growth factor-beta 1(TGF-β1),alpha-smooth muscle actin(α-SMA),and hepatocyte growth factor(HGF)in renal tissue.Results Compared with the control group,the model group had significant increases in serum creatinine(SCr),blood urea nitrogen(BUN),and the protein and mRNA expression levels of TGF-β1,HGF,andα-SMA in renal tissue(P<0.05);compared with the model group,the Uremic Clearance Granule group,losartan group,and Huazhuo Jiedu group had significant reductions in SCr,BUN,and the protein and mRNA expression levels of TGF-β1 andα-SMA in renal tissue(P<0.05),as well as significant increases in the protein and mRNA expression levels of HGF in renal tissue(P<0.05).The Uremic Clearance Granule group,losartan group,and Huazhuo Jiedu group showed varying degrees of improvement in renal fibrosis.Conclusion Huazhuo Jiedu Prescription can effectively improve the renal function of CRF rats,possibly by inhibiting the expression of TGF-β1 and upregulating the expression of HGF in renal tissue,thereby inhibiting the expression ofα-SMA.
