摘 要:为了解河北省鸭圆环病毒(DuCV)的感染情况,本试验对河北省鸭场送检的48份以脱毛、发育不良为特征,临床诊断为DuCV感染的病死鸭组织进行常规PCR检测,并从阳性样本中选取2株DuCV扩增全基因序列,通过DNASTAR和MEGA 7.0进行序列比对及遗传进化分析。结果显示:48份样本中,常规PCR检测出35份DuCV阳性;选取的2株DuCV(登录号分别为MW255979和MW255980)均属于DuCV-1基因型,全基因序列的同源性为97.9%,与14株国内外参考株序列同源性为82.9%~98.7%;其中MW255979与中国广西株MK814578核苷酸同源性最高,为98.7%,与中国山东株EU022374和中国广西株JX241045核苷酸同源性最低,为83.0%;MW255980与韩国株JQ740360核苷酸同源性最高,为98.6%,与中国山东株EU022374核苷酸同源性最低,为82.9%。结果表明,本试验获得的2株DuCV全基因序列与近年来我国大陆地区临床所获得的DuCV-1型毒株相比,基因型未发生明显突变,本结果充实了河北省DuCV感染情况的资料库,为进一步研究DuCV基因提供了数据支持。In order to clarify the infection situation of duck circovirus(DuCV)in Hebei Province,this study utilized conventional PCR to assess 48 tissues from dead ducks with clinically diagnosed DuCV infection,which were characterized by hair loss and dysplasia,and selected two DuCV strains from the PCR positive samples for complete gene sequencing and sequence alignment and genetic evolution analysis by DNASTAR and MEGA 7.0.The results showed that 35 of the 48 samples were PCR positive for DuCV;the 2 selected DuCV strains(with accession numbers MW255979 and MW255980,respectively)belonged to the DuCV-1 genotype,sharing nucleotide sequence homology of 97.9%between them,and 82.9%~98.7%with the 14 domestic and foreign reference strains;MW255979 had the highest nucleotide homology with Guangxi strain MK814578(98.7%),and the lowest nucleotide homology with Shandong strain EU022374 and Guangxi strain JX241045(83.0%);MW255980 had the highest nucleotide homology with Korean strain JQ740360(98.6%),and the lowest homology with EU022374(82.9%).Thus,the whole genome sequences of the two DuCV strains obtained in this study were not significantly mutated compared with those of the DuCV-1 strains obtained clinically in China's Mainland in recent years.These results enriched the database of DuCV infection in Hebei Province,and provided data support for further genetic study of DuCV.
