阿司匹林对高脂乳剂致大鼠骨丢失的骨形态计量学研究

摘  要:本试验用高脂乳剂建立高脂血症致大鼠骨丢失模型,通过骨形态计量学指标观察阿司匹林对大鼠胫骨上段(PTM)和中段(Tx)骨丢失的防治作用。选取3月龄SPF级SD大鼠30只,雌雄各半,随机分为生理盐水(NS)组、高脂乳剂(HFE)组和阿司匹林(AP)组,每组10只。给药20周后取材,分离血清,检测总胆固醇(TC)、甘油三酯(TG)和高密度脂蛋白胆固醇(HDL-C)含量,并取胫骨上段、中段检测骨组织形态计量学参数。结果显示,与NS组比较,HFE组大鼠TC极显著升高、HDL-C极显著降低(P<0.01);胫骨上段的成骨细胞周长百分率(%Ob.S.Pm)显著减少(P<0.05),骨小梁数量(Tb.N)和骨小梁面积百分数(%Tb.Ar)均极显著减少(P<0.01),而骨小梁分离度(Tb.Sp)、每毫米破骨细胞数量(Oc.N/mm)、每平方毫米破骨细胞数量(Oc.N/mm 2)和破骨细胞周长百分率(%Oc.S.Pm)均极显著增加(P<0.01);胫骨中段的皮质骨面积百分数(%Ct.Ar)极显著减少(P<0.01),骨髓腔面积百分数(%Ma.Ar)极显著增加(P<0.01)。阿司匹林可使高脂大鼠TC、TG显著下降,HDL-C显著升高(P<0.05);胫骨上段骨小梁宽度(Tb.Th)显著增加(P<0.05),Tb.N、%Tb.Ar和%Ob.S.Pm均极显著增加(P<0.01),而Tb.Sp、Oc.N/mm、Oc.N/mm 2和%Oc.S.Pm均极显著减少(P<0.01);胫骨中段的%Ct.Ar显著增加(P<0.05),%Ma.Ar极显著减少(P<0.01)。结果表明,长期给予大鼠高脂乳剂灌胃会导致胫骨上段和中段的骨丢失,阿司匹林能通过促进骨生成有效对抗骨丢失。To study the effect of aspirin on the prevention and treatment of bone loss of the longitudinal proximal tibial metaphyseal(PTM)and tibial shaft(Tx)by using bone histomorphometry through establishing a bone loss rat model induced by high-fat emulsion.Thirty 3-months-old specific-pathogen free(SPF)SD rats were randomly divided into normal saline(NS)group,high fat emulsion(HFE)group,aspirin(AP)group.Rats were sacrificed after 20 weeks,serum total cholesterol(TC),triglyceride(TG),and high-density lipoprotein cholesterol(HDL-C)levels were measured,and undecalcified PTM and Tx sections were performed and used for bone histomorphometric analysis.Compared with NS group,serum TC increased and HDL-C decreased significantly in HFE group(P<0.01).In PTM of high fat emulsion treated rats,osteoblast perimeter percentage(%Ob.S.Pm)decreased(P<0.05),trabecular bone number(Tb.N)and trabecular bone area percentage(%Tb.Ar)decreased significantly(P<0.01),while trabecular bone separation(Tb.Sp),osteoclast number per mm(Oc.N/mm),osteoclast number per mm 2(Oc.N/mm 2),and osteoclast perimeter percentage(%Oc.S.Pm)increased significantly(P<0.01).In Tx of high fat emulsion treated rats,cortical bone area percentage(%Ct.Ar)decreased significantly(P<0.01),while bone marrow cavity area percentage(%Ma.Ar)increased significantly(P<0.01).Compared with HFE group,serum TC,TG decreased and HDL-C increased(P<0.05).In PTM of aspirin treated rats,trabecular bone thickness(Tb.Th)increased(P<0.05),Tb.N,%Tb.Ar,and%Ob.S.Pm increased significantly(P<0.01),while Tb.Sp,Oc.N/mm,Oc.N/mm 2 and%Oc.S.Pm decreased significantly(P<0.01).In Tx of aspirin treated rats,%Ct.Ar increased(P<0.05),while%Ma.Ar decreased significantly(P<0.01).Thus,long-term high-fat emulsion diets may cause loss of PTM and Tx in rats,and aspirin can effectively prevent those changes through stimulating bone formation.
