摘  要:目的探究鳖甲-莪术药对对TGF-β1诱导的MDA-MB-231细胞侵袭转移能力的影响及细胞上皮间质转化(EMT)的作用及机制。方法将含药血清体外培养的MDA-MB-231细胞设空白组、单诱导组、鳖甲组、莪术组、鳖甲-莪术组以及紫杉醇组,采用CCK-8法、划痕实验、Transwell小室实验分别检测细胞增殖、迁移和侵袭能力,Western blot法和RT-qPCR法分别检测E钙黏蛋白(E-cadherin)、波形蛋白(vimentin)、N钙黏蛋白(N-cadherin)、Wnt3a、β-连环蛋白(β-catenin)蛋白及mRNA表达。结果与单诱导组相比,各实验组细胞增殖抑制率增加、细胞迁移率降低、细胞迁徙数目减少(P<0.05),提示各实验组细胞增殖、迁移以及侵袭能力均降低;组间比较,鳖甲-莪术组对MDA-MB-231细胞的增殖抑制率高于鳖甲组和莪术组(P<0.05);与单诱导组相比,各实验组的E-cadherin蛋白及mRNA表达均上升,Vimentin、N-cadherin、Wnt3a、β-catenin蛋白及mRNA表达均下降(P<0.05);组间比较,鳖甲-莪术组E-cadherin蛋白及mRNA表达最高,Vimentin、N-cadherin、Wnt3a、β-catenin蛋白及mRNA表达低于鳖甲组、莪术组(P<0.05)。结论鳖甲-莪术药对能抑制TGF-β1诱导的MDA-MB-231细胞侵袭转移能力及其EMT进程,其机制可能与调节Wnt/β-catenin信号通路中某些蛋白及基因水平有关。Objective To investigate the effect of Trionycis Carapax-Zedoary Turmeric drug pair on invasion and metastasis of MDA-MB-231 cells induced by TGF-β1 and its epithelial-mesenchymal transition(EMT)effect and mechanism.Methods MDA-MB-231 cells were cultured in vitro with drug-containing serum.Divided into blank group,single induction group,trionycis carapax group,zedoary turmeric group,trionycis carapax-zedoary turmeric drug pair group,and paclitaxel group.Cell proliferation,migration and invasion were detected by CCK-8 assay,scratch assay,and Transwell chamber assay respectively.E-cadherin,vimentin,N-cadherin,Wnt3a,β-catenin protein and mRNA expression were detected by Western blot and RT-qPCR respectively.Results Compared with single induction group,cell proliferation inhibition was increased,migration was decreased and number of cell invasion reduced in each experimental group(P<0.05).The study indicated cell proliferation,migration and invasion of all experimental groups decreased.Compared among experimental groups,the inhibitory effect of Trionycis Carapax-Zedoary Turmeric drug pair group was better than that of Trionycis Carapax and Zedoary Turmeric alone(P<0.05).Compared with single induction group,the expression of E-cadherin protein and mRNA increased,and the expression of Vimentin,N-cadherin,Wnt3a,β-catenin protein and mRNA decreased in each experimental group(P<0.05).Compared among experimental groups,the expression of E-cadherin protein and mRNA was the highest in the Trionycis Carapax-Zedoary Turmeric drug pair group,and the expression of Vimentin,N-cadherin,Wnt3a,β-catenin protein and mRNA was lower than that of Trionycis Carapax and Zedoary Turmeric groups(P<0.05).Conclusion Trionycis Carapax-Zedoary Turmeric drug pair can inhibit the invasion and metastasis of MDA-MB-231 cells induced by TGF-β1 as well as EMT process,and the effect is better than that of trionycis carapax and zedoary turmeric alone.Possible mechanism can be associated with certain protein and genetic level regulating Wnt/β-cate
