摘  要:为探究串晶(shish-kebab,SK)结构片晶密度对成纤维细胞行为的影响及其机制,结合静电纺丝技术和溶液孵育结晶法制备了表面光滑和具有不同片晶密度的串晶结构纤维支架,通过分析表面形貌、力学性能及体外细胞试验结果,探究不同拓扑结构对人皮肤成纤维细胞的形态及其增殖和基因表达的影响。结果表明,低密度的串晶结构(粗糙度为(0.85±0.10)μm)可以改善纤维支架的表面和力学性能,增大细胞铺展面积,并减少转化生长因子(TGF-β_(1))和α-平滑肌肌动蛋白(α-SMA)的表达,加快组织修复,减少瘢痕,在伤口修复中具有应用潜力。The smooth and series shish-kebab(SK)fiber scaffolds with different kebab densities were prepared by electrospinning and solution incubation methods to investigate the effect of kebab density of SK structure on the behavior of fibroblasts and its mechanism.The effects of fiber scaffolds with different surface topologies on the morphology,proliferation and gene expression of human foreskin fibroblasts were explored by analyzing the surface morphology,roughness,mechanical properties and the results of in vitro cell tests.The results show that the low-density SK structure(roughness:(0.85±0.10)μm)can improve the surface and mechanical properties of fiber scaffolds,increase cell spread area,reduce the expression of transforming growth factor-beta1(TGF-β_(1))andα-smooth muscle actin(α-SMA),accelerate wound healing and reduce scar tissues,having potential applications in wound healing.
