摘 要:为探究木薯两性花和正常花基因组DNA的甲基化模式以及甲基化水平差异,并找到木薯两性花与正常花之间的差异甲基化基因,本实验采用全基因组甲基化测序技术(WGBS)对木薯两性花(WLa)和正常花(CKa)的基因组DNA甲基化水平进行分析比较。结果表明,木薯花内的DNA甲基化模式主要有mCG、mCHG、mCHH三种类型,CG位点的平均甲基化水平最高。WLa处于全甲基化状态(ML100)的mCG和mCHG均多于CKa,而mCHH则略低于CK。对木薯两性花和正常花进行差异甲基化(DMR)鉴定研究,共检测到14987个DMR和5697个DMGs。另外,基因本体(GO)分析显示,基因高度富集在催化活性、结合、代谢等过程中。相关信号通路(KEGG)分析则表明。基因主要富集到赖氨酸生物合成、ABC转运蛋白体等代谢通路中。In order to explore the methylation patterns of bisexual flowers of cassva and find the differential methylated genes between cassava bisexual flowers and normal flowers,we use the whole genome bisulfite sequencing technology(WGBS).The gene methylation levels and differential methylation regions of cassava bisexual flowers(WLa)and normal flowers(CKa)were analyzed.The results indicated that there were three main types of DNA methylation in cassava flowers:mCG,mCHG,and mCHH.The mean methylation level was highest at the CG locus.mCG and mCHG in the fully methylated state(ML100)were both higher in WLa than in CKa,while mCHH was slightly lower than in CK.14987 DMRs and 5697 DMGs were detected in a differential methylation(DMR)identification study of cassava bisexual and normal flowers.In addition,gene ontology(GO)analysis shows that genes are highly enriched in processes such as catalytic activity,binding,and metabolism.Related signal path way(KEGG)analysis shows that.Genes are mainly enriched in metabolic pathways such as lysine biosynthesis and ABC transporters.
