云南白灵芝菌株分子鉴定及SSR遗传多样性分析

摘　　要：为了在分子水平上明确云南白灵芝的种属地位和遗传差异,本实验利用rDNA-ITS序列比对和SSR分子标记法,对云南白灵芝菌株进行分子鉴定和遗传多样性分析。结果显示,15个菌株的ITS测序在种的水平上鉴定为白肉灵芝(Ganoderma leucocontextum);SSR-PCR筛选出17对扩增稳定、多态性较高的引物,参试菌株共检测出101个等位基因位点,等位变异数范围为3~10个,平均每对引物等位基因数目为5.94个;Nei’s遗传多样性指数变化范围为0.127~0.330,平均为0.223;香农指数在0.231~0.504之间,平均值为0.365。经遗传多样性分析,各样本间的相似水平在0.40~0.75之间。研究表明各菌株间存在较大的遗传差异,为云南白灵芝的菌种鉴定和新品种测试提供依据。In order to clarify the species status and genetic diversity of Ganoderma leucocontextum at molecular level,rDNA-ITS sequence alignment and SSR markers were used to identify and analyze the genetic diversity of Ganoderma strains from Yunnan.The ITS regions analysis of the 15 strains indicated that the tested strains were G.leucocontextum.17 pairs of SSR primers were selected according to the stability and difference of amplification bands.A total of 101 alleles were detected.For each pair of primers,the alleles detected ranged 3 from 10 with an average of 5.94,the Nei’s genetic diversity index ranged 0.127 from 0.330 with an average of 0.223,the shannon’s index ranged 0.231 from 0.504 with an average of 0.365.The genetic diversity analysis showed that the similarity between the samples ranged from 0.40 to 0.75.The results showed that there were great genetic differences among the strains,which provided the basis for the identification of Ganoderma strains from Yunnan and the testing of new varieties.
