摘  要:目的 探讨铁皮石斛对小鼠的急性毒性和遗传毒性。方法 将铁皮石斛粉碎制成粉末,作为受试物。采用限量法进行急性经口毒性试验,选用20只小鼠,雌雄各半,累积染毒剂量为12.5 g/(kg·bw)。采用平板掺入法进行细菌回复突变试验,设5 000、1 000、200、40、8 μg/皿共5个剂量组,在有或无代谢活化系统(S9)的条件下计数每皿回复突变菌落数。行哺乳动物红细胞微核试验,设2.50、1.25、0.63 g/(kg·bw)剂量组,计数出现微核的嗜多染红细胞并计算微核率。行小鼠精原细胞染色体畸变试验,设5.00、2.50、1.25 g/(kg·bw)剂量组,观察染色体结构畸变类型并计算畸变率。3个遗传毒性试验均另设置阴性对照组和阳性对照组。结果 铁皮石斛对雌雄小鼠的LD均>12.5 g/kg·bw。各组细菌回复突变试验、哺乳动物红细胞微核试验和小鼠精原细胞染色体畸变试验均为阴性。结论 在本研究条件下,未发现铁皮石斛有急性毒性反应和遗传毒性。Objective To study the acute toxicity and genotoxicity of Dendrobium officinale in mice. Methods The dry strip of Dendrobium officinale was pulverized into powder and used as the test.For acute oral toxicity test, male and female mice(10 in each group) were orally gavaged with a cumulative dose of 12.5 g/(kg·bw).In the bacterial recovery mutation test, the plate incorporation method was used to set 5 dose groups including 5 000,1 000,200,40 and 8 μg/dish, and the number of mutant colonies per dish was counted with or without metabolic activation system(S9).Mammalian erythrocyte micronucleus test was set as 2.50,1.25 and 0.63 g/(kg·bw) dose groups.The polychromatic erythrocytes with micronucleus were counted and the micronucleus rate was calculated.Mice spermatogonial chromosome aberration test was set as 5.00,2.50 and 1.25 g/(kg·bw) dose groups.The types of chromosome structural aberration were observed and the aberration rate was calculated.Negative control group and positive control group were also set up in the 3 genotoxicity tests. Results The LDof Dendrobium officinale to male and female mice was greater than 12.5 g/(kg·bw).Bacterial recovery mutation test, mammalian erythrocyte micronucleus test and mouse spermatogonial chromosome aberration test were all negative. Conclusion No acute toxicity and genotoxicity of Dendrobium officinale are found in this study.
