摘 要:目的 探究不同生境下不同生长期马蓝叶指纹图谱和化学特征,为马蓝的栽培过程中光强管理和品质评价提供方法和参考。方法 采集11批不同生境(桉树下、稻田半阳生、稻田全阴生、稻田全日照、枇杷树下)和不同生长期马蓝叶,采用HPLC法建立11批马蓝叶指纹图谱,同时测定马蓝叶主要成分吲哚苷、色胺酮、靛蓝和靛玉红的含量,结合相似度评价、聚类分析和主成分分析的化学计量学方法对不同生境和生长期马蓝叶的指纹图谱和化学特征进行分析。结果 建立11批马蓝叶的指纹图谱,共标定8个峰,指认4个峰,指纹图谱相似度在0.295~0.806,生长后期的马蓝叶非极性成分增加;聚类分析和主成分分析一致,分为4大类,稻田全日照马蓝叶与其他生境马蓝叶显著不同。含量分析结果表明马蓝叶中吲哚苷、靛蓝和靛玉红含量在生长后期增加,而色胺酮降低。光照对马蓝叶4种成分吲哚苷、色胺酮、靛蓝和靛玉红有一定影响,全日照和部分光照均有利于马蓝叶吲哚苷、靛蓝和靛玉红的积累,部分光照有利于马蓝叶色胺酮的积累。结论 马蓝叶指纹图谱和化学成分受不同生境和不同生长期的影响,稻田和树荫下均适合马蓝的栽培,部分光照更有利于马蓝叶活性成分的积累。本研究所建立的指纹图谱、多指标定量结合化学计量学分析方法为马蓝的栽培光照强度管理和质量评价提供方法参考。Objective To provide methods and references for light intensity management and quality evaluation of Baphicacanthus cusia,the fingerprint and chemical characteristics of Baphicacanthus cusia leaves at different growth stages under different habitats were studied.Methods Eleven batches of different habitats(under eucalyptus, half-sunny in paddy field, full-sunny in paddy field, full sunlight in paddy field, and under loquat tree) and different growth stages of Baphicacanthus cusia leaves were collected.The 11 batches of Baphicacanthus cusia leaves fingerprints were established by HPLC method, and the contents of the main effective components indole glycosides, tryptanthrin, indigo and indirubin were analyzed, combined with similarity evaluation, cluster analysis(CA) and principal component analysis(PCA) method for samples analysis of the fingerprint and chemical characteristics.Results The fingerprints of 11 batches of Baphicacanthus cusia leaves were established, and 8 peaks were calibrated, and 4 peaks were identified.The similarity of the fingerprints was 0.295-0.806.The non-polar components of samples with longer growth period increased.The CA and PCA analysis were consistent, and samples were divided into 4 categories.The Baphicacanthus cusia leaves of the paddy field were significantly different from those of other habitats.The results of the content analysis showed that the contents of indole glycosides, indigo and indirubin in most of samples increased in the late growth stage, while tryptophanone decreased.Light had a certain effect on the indole glycosides, tryptanthrin, indigo and indirubin of Baphicacanthus cusia leaves.Full daylight and partial light were conducive to the accumulation of indole glycosides, indigo and indirubin, and partial light was conducive to the accumulation of tryptanthrin.Conclusion The fingerprint and chemical composition of Baphicacanthus cusia leaves are affected by different habitats and different growth periods.Both rice fields and shades are suitable for the cultivation of
