摘 要:目的:分析艾塞那肽介导线粒体凋亡途径保护大鼠心肌缺血再灌注损伤(MIRI)的作用及其机制。方法:采用随机数表法将36只大鼠分为假手术组、MIRI组和艾塞那肽组(10μg/kg,皮下注射),每组各12只。恢复血流灌注120 min后,检测各组大鼠心肌梗死面积和心肌组织损伤情况;检测各组大鼠血清心肌酶水平和氧化应激水平,蛋白印迹法检测各组大鼠心肌组织Bcl-2基因、Bax、Cleaved Caspase-3、胞浆细胞色素C(Cyt-c)和线粒体Cyt-c蛋白表达情况。结果:与假手术组相比,MIRI组大鼠的心肌梗死面积和细胞凋亡率升高(P<0.05),血清心肌酶和MDA表达水平升高(P<0.05),SOD和GSH-px表达水平降低(P<0.05),心肌组织Bax/Bcl-2、Cleaved Caspase-3和胞浆Cyt-c蛋白表达水平升高(P<0.05),线粒体Cyt-c蛋白表达降低(P<0.05);与MIRI组相比,艾塞那肽组大鼠的心肌梗死面积和细胞凋亡率降低(P<0.05),血清心肌酶和MDA表达水平降低(P<0.05),SOD和GSH-px表达水平升高(P<0.05),心肌组织Bax/Bcl-2、Cleaved Caspase-3和胞浆Cyt-c蛋白表达水平降低(P<0.05),线粒体Cyt-c蛋白表达水平升高(P<0.05)。结论:艾塞那肽可抑制MIRI大鼠线粒体凋亡途径介导的心肌组织细胞凋亡,减轻心肌损伤。Objective:To analyze the roles and mechanism of exenatide on protecting from myocardial ischemia-reperfusion injury(MIRI) in rats by mediating mitochondrial apoptosis.Methods:A total of 36 rats were divided into sham operation group, MIRI group and exenatide group(10 μg/kg, subcutaneous injection),12 cases in each group.After reperfusion for 120 min, area of myocardial infarction and myocardial tissue damage were detected.The levels of serum myocardial enzyme and oxidative stress were detected.The expression of B-cell lymphoma/leukemia-2(Bcl-2) gene, Bcl-associated xprotein(Bax),Cleaved Caspase-3,cytochrome C(Cyt-c) and mitochondrial Cyt-c protein were detected by Western blot.Results:Compared with sham operation group, the myocardial infarction area and apoptosis rate of myocardial cells in MIRI group were increased(P<0.05),levels of serum myocardial enzyme and malondialdehyde(MDA) were increased(P<0.05),superoxide dismutase(SOD) and glutathione peroxidase(GSH-px) levels were decreased(P<0.05),expression of Bax/Bcl-2,Cleaved Caspase-3 and Cyt-c protein in myocardial tissue were increased(P<0.05),and mitochondrial Cyt-c was decreased(P<0.05).Compared with MIRI group, the myocardial infarction area and apoptosis rate of myocardial cells in exenatide group were decreased(P<0.05),levels of serum myocardial enzyme and MDA were decreased(P<0.05),SOD and GSH-px levels were increased(P<0.05),expression of Bax/Bcl-2,Cleaved Caspase-3 and Cyt-c protein in myocardial tissue were decreased(P<0.05),and mitochondrial Cyt-c was increased(P<0.05).Conclusion:Exenatide can inhibit apoptosis of myocardial tissue cells mediated by mitochondrial apoptosis pathway in MIRI rats, and reduce myocardial tissue damage.
