摘 要:[目的]本文旨在探讨轮作对设施草莓土壤细菌群落多样性的影响。[方法]设置草莓-玉米、草莓-大豆、草莓-西瓜、草莓-苜蓿4个轮作处理,以草莓连作为对照,利用Illumina Miseq高通量测序技术,分析了不同轮作模式对设施草莓根际土壤细菌的群落结构、丰富度及多样性指数变化特征。[结果]所有土壤样本中共获得643898个有效序列。变形菌门(Proteobacteria)、绿弯菌门(Chloroflexi)、放线菌门(Actinobacteria)、芽单胞菌门(Gemmatimonadetes)和酸杆菌门(Acidobacteria)是草莓根际土壤细菌的优势菌门。不同作物与草莓轮作能够改变草莓根际土壤细菌群落,采取草莓-苜蓿轮作的种植模式在门、纲、目、科的分类水平上增加了土壤细菌的丰富度,且表现出了土壤中最多的细菌群落;而草莓连作的土壤细菌的丰富度最低,草莓-大豆轮作模式的土壤细菌的群落多样性最低。[结论]本研究有助于从微生物的作用机制分析比较轮作对设施草莓的影响,为设施草莓的生产提供理论依据。[Objective]Present study was designed to explore the effects of crop rotation on the bacterial community diversity of strawberry soil in the facilities. [Methods]Four crop rotational models were set up,including strawberry/maize,strawberry/soybean,strawberry/watermelon,and strawberry/alfalfa. The fields with continuous planting of strawberry were served as control. Illumina MiSeq high-throughput sequencing technology analysis was used to monitor the effects of different rotation patterns on the community structure,bacterial richness and the diversity index of strawberry rhizosphere soil. [Results]In this experiment,MiSeq sequencing analysis results showed that a total of 643 898 OTUs were obtained from all soil samples. Proteobacteria,Chloroflexi,Actinobacteria,Gemmatimonadetes and Acidobacteria were the dominant phyla of strawberry rhizosphere soil bacteria. Different crops/strawberry rotation changed the bacterial communities of strawberry rhizosphere soil. The planting mode of strawberry/alfalfa rotation increased the richness of soil bacteria at the classification level of phylum,class,order and family,and showed the most abundant soil bacterial community. The richness of soil bacteria in control group was the lowest,and the community diversity of soil bacteria in strawberry/soybean rotation mode was the lowest. [Conclusion]The above research is of importance to analyze and compare the effects of crop rotation on strawberry production in facilities from the mechanism of action of microorganisms,and provides theoretical basis for the production of facility strawberries.
