摘  要:葡萄霜霉病是一种世界性病害,对葡萄生产影响极大,是葡萄栽培中主要病害之一。基于非靶向代谢组学研究技术,分析鉴定对照组与霜霉病组代谢物含量的变化,采用正交偏最小二乘判别分析并结合t检验的结果,筛选出造成健康和发病的葡萄叶片显著差异的20种主要代谢物质。其中,霜霉病处理组差异代谢物含量显著上升(P<0.05)的物质主要分为4类:氨基酸类、核酸代谢类、植物激素类和其他化合物;显著下降(P<0.05)的物质分6类:糖类、羧酸类、甾类、二萜醇类、三萜类和其他化合物。KEGG通路富集分析发现差异代谢物主要富集在氨酰tRNA生物合成,ABC转运蛋白,戊糖磷酸途径,二苯乙烯类、二芳基庚烷类和姜辣素生物合成,缬氨酸、亮氨酸和异亮氨酸生物合成和植物激素信号转导6条代谢通路中。由此可知,葡萄霜霉病侵染导致了葡萄叶片糖、核酸、氨基酸等相关代谢物的显著变化,相关代谢途径可能是霜霉菌侵染葡萄的主要机制。Grape downy mildew is one of the main diseases in grape cultivation worldwide,which has a great impact on grape production.Based on the nontargeted metabonomics research technology,the changes of meta-bolites in the control group and the downy mildew group were analyzed.Twenty main metabolites causing significant differences among the healthy and the diseased grape leaves were selected by orthogonal partial least squares discriminant analysis and t-test,among which the substances with content of differential metabolites significantly(P<0.05)increased in downy mildew treatment group were mainly divided into four categories:amino acids(isoleucine,tryptophan,proline),nucleic acid metabolism(trigonelline),plant hormones(salicylic acid,indoleacetic acid),the other compounds(pterostilbene,epsilon-viniferin).The substances with content of differential metabolites significantly(P<0.05)decreased were divided into six categories:sugars(ribose),carboxylic acids(7,10-heptadecanedioic acid),steroids(4 a-methylzymosterol),and diterpenoid alcohols(sclareol),triterpenoid(lupenone,lucidenic acid C)and other compounds(rifabutin,3-hydroxy-beta-ionone,sandosapogenol,goshonoside F 5,fasciculol E,25-acetylvulgaroside).The KEGG pathway enrichment analysis showed that the differential metabolites were mainly enriched in six metabolic pathways,such as aminoacyl tRNA biosynthesis,ABC transporter,pentose phosphate pathway,stilbene,stilbenoid,diarylheptanoid and gingerol biosynthesis,valine,leucine and isoleucine biosynthesis,and plant hormone signal transduction.Therefore,the infection of grape downy mildew caused significant changes in glucose,nucleic acid,amino acid and other related metabolites in grape leaves,which might be the main mechanism of grape downy mildew involved in infection.
