摘  要:目的:分析自体富血小板凝胶(APG)联合脂肪干细胞(ADSCs)对糖尿病足(DF)大鼠lncRNA的影响。方法:2021年6-9月,选取30只SPF级C57BL/6大鼠,适应性喂养1周后随机分为正常组、对照组、治疗组,各10只,正常组自由饮水不建模,其余两组建立DF模型,其中对照组注射磷酸缓冲盐溶液(PBS)于DF大鼠溃疡处,治疗组注射APG+ADSCs于溃疡处,连续治疗14 d后测定大鼠血糖、炎症因子[白介素-6(IL-6)、C反应蛋白(CRP)、肿瘤坏死因子-α(TNF-α)]水平与血管内皮生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)水平,同时收集溃疡组织进行lncRNA芯片分析,测定lncRNA表达谱变化。结果:治疗14 d后,正常组血糖与治疗前比较,差异无统计学意义(P>0.05),对照组血糖水平高于治疗前,治疗组血糖水平低于治疗前,且治疗组低于对照组(P<0.05);治疗14 d后,治疗组IL-6、CRP、TNF-α均低于对照组,而VEGF、bFGF均高于对照组(P<0.05)。与正常组比较,对照组存在1627个显著差异的lncRNA,治疗组存在1896个显著差异的lncRNA,与治疗组比较,对照组存在2576个显著差异的lncRNA。将靶基因代入差异mRNA数据,筛选得到DF相关的靶基因有8个,分别为DLX6-AS1、CHI3L1、CAMTA1、STRADB、ARAP1-AS1、ARAP1-AS2、PVT1、ROR。结论:APG联合ADSCs治疗DF的机制可能是通过改变lncRNA的表达,影响炎症因子与血管生成因子水平而发挥作用。Objective:To analyze the effect of autologous platelet-rich gel(APG)combined with adipose-derived stem cells(ADSCs)on lncRNA in rats with diabetic foot(DF).Method:From June to September 2021,30 SPF C57BL/6 rats were selected,and randomly divided into the normal group,the control group and the treatment group after 1 week of adaptive feeding,with 10 rats in each group.Rats in the normal group drank freely,while the other two groups were used for establishing DF models.Rats in the control group were injected with Phosphate Buffered Saline(PBS)at the ulcer site,while those in the treatment group were injected with APG and ADSCs at the ulcer site.The levels of blood glucose,inflammatory factors[interleukin-6(IL-6),C reactive protein(CRP)and tumor necrosis factor-α(TNF-α)],vascular endothelial growth factor(VEGF)and basic fibroblast growth factor(bFGF)were measured after 14 days of continuous treatment.The ulcer tissues were collected for lncRNA chip analysis,and the changes of lncRNA expression profile were determined.Result:After 14 days of treatment,the difference in blood glucose of the normal group was not statistically significant as compared with that before treatment(P>0.05),the blood glucose level of the control group was higher than that before treatment,the blood glucose level of the treatment group was lower than that before treatment,and the blood glucose level of the treatment group was lower than that of the control group(P<0.05).After 14 days of treatment,the levels of IL-6,CRP,TNF-αin the treatment group were lower than those of the control group,while the levels of VEGF and bFGF were higher than those of the control group(P<0.05).Compared with the normal group,there were 1627 significantly different lncRNA in the control group and 1896 significantly different lncRNA in the treatment group.Compared with the treatment group,there were 2576 significantly different lncRNA in the control group.By substituting target genes into the differential mRNA data,we screened out eight DF-related target genes,whi
