摘  要:目的研究祛痰法代表方三子养亲汤对大鼠哮喘模型气道重构的影响及机制。方法将35只大鼠分为空白组、哮喘模型组、三子养亲汤组、三子养亲汤+发动蛋白抑制剂(mitochondrial division inhibitor,Mdivi-1)组和Mdivi-1组共5组,采取鸡卵清白蛋白(ovalbumin,OVA)联合氢氧化铝致敏,OVA激发建立哮喘大鼠模型,OVA激发前灌胃给药,末次灌药后取肺组织标本,观察大鼠肺组织炎症及黏液状态,电子显微镜观察线粒体自噬情况;Western blot法检测肺组织中程序性死亡受体-1(Beclin1)紫外线抵抗相关基因(UVRAG)蛋白表达量,qPCR测定肺组织Beclin1、UVRAG mRNA表达。结果与空白组比较,哮喘模型组大鼠肺组织中Beclin1、UVRAG蛋白、mRNA均显著升高(P<0.01);三子养亲汤组肺组织中炎症浸润减少,杯状细胞增殖减少,电镜下线粒体结构正常,较少自噬体形成,Beclin1、UVRAG蛋白表达明显降低(P<0.01),而此种效果能被Mdivi-1逆转。结论三子养亲汤能改善哮喘大鼠气道重构,可能与其下调Beclin1、UVRAG表达有关。Objective To investigate the effect and mechanism of Sanzi Yangqin Decoction on airway remodeling in asthmatic rats.Methods According to the random number table method,35male rats were divided into control group,asthma model group,Sanzi Yangqin Decoction group,Sanzi Yangqin Decoction group+Mdivi-1 group and Mdivi-1 group.The study performed gavaging before OVA challenge,once a day for 15 consecutive days,and 24 hours after the last challenge,with hematoxylin-eosin(HE)staining and periodic acid Schiff(PAS)staining to observe the inflammation and mucus status of rat lung tissue.We used electron microscope to observe the mitochondrial autophagy of rat lung tissue,Western blot method to detect the programmed death receptor-1 in lung tissue(Beclin1)Ultraviolet resistance-related gene(UVRAG)protein expression qPCR to determine the expression of Beclin1,UVRAG mRNA in lung tissue.Results Compared with the control group,Beclin1,UVRAG protein,mRNA and pre-mRNA in the lung tissue of the asthma model group were significantly increased(P<0.01).Compared with the model group,in the Sanzi Yangqin Decoction group,the inflammatory infiltration and goblet cell proliferation were reduced in the lung tissues of the Sanzi Yangqin Decoction group,the structure of mitochondria under the electron microscope was normal,and there was less autophagosome formation,and the protein expression of Beclin1 and UVRAG was significantly reduced(P<0.01),and this effect can be reversed by Mdivi-1.Conclusion Sanzi Yangqin Decoction can improve airway remodeling in asthmatic rats,which may be related to the down-regulation of Beclin1 and UVRAG expression to restore mitochondrial autophagy homeostasis.
