摘 要:目的 运用Plackett-Burnman(PB)试验设计和Box-Behnken设计(BBD)-效应面法(RSM)探讨和优化三颗针中盐酸小檗碱闪式提取工艺。方法 以盐酸小檗碱的转移率为响应值,PB试验设计筛选出3个显著性因素,最陡爬坡试验逼近响应值最优区域,BBD-RSM试验设计对显著性因素进行效应面优化,得到影响盐酸小檗碱得率的二阶数学模型,进而得出最佳工艺条件。结果 闪式提取盐酸小檗碱最佳工艺条件为:液料比为25 ml/g,乙醇浓度为70%,提取时间为30 s。结论 优化后工艺操作具有可行性,可为三颗针的开发利用提供支持。Objective To optimize the fiash extraction technology of berberine hydrochloride from Berberidis Radix by Plackett-Burnman(PB) design and Box-Behnken design(BBD)-response surface methodology(RSM).Methods Taking the transfer rate of berberine hydrochloride as the response value,three significant factors were screened by PB design.The optimal region of response value was obtained by steepest climbing test,and BBD-RSM was used to carry out the response surface optimization of the significant factors to obtain a two-order mathematical model affecting the yield of berberine hydrochloride,as well as the optimum process condition.Results The best condition for fiush extraction of berberine hydrochloride from Berberidis Radix was as follows:the ratio of liquid to material was 25 ml/g,the ethanol concentration was 70 % and the extraction time was 30 s.Conclusion The optimized fiush extraction process of berberine hydrochloride from Berberidis Radix is feasible,which can provide supply for the development and utilization of Berberidis Radix.
