摘 要:本文探究了一种剪切活化纳米颗粒药物靶向递送策略,可用于辅助治疗主动脉缩窄和主动脉瘤。基于理想化的主动脉缩窄和主动脉瘤的三维几何模型,考虑了脉动血流与主动脉壁面的流固耦合作用,利用病变主动脉内局部独特的血流动力学环境实现纳米颗粒药物的靶向输运,搭载纳米颗粒药物的微米载体将被局部异常的剪切率激活进而靶向释放纳米颗粒药物。本文初步论证了剪切活化纳米颗粒药物靶向输运策略在主动脉病变治疗中的可行性,研究结果可以为设计药物输运系统和开发新型治疗方法提供理论依据。In this study, we explore a shear-activated targeted nanoparticle drug delivery strategy for adjuvant treatment of aortic coarctation and aortic aneurysm. The fluid-structure interaction between pulsatile blood flow and the aortic wall is considered based on the idealized three-dimensional geometric model of aortic coarctation and aortic aneurysm. The local unique hemodynamic environment in the diseased aorta is used to achieve nanoparticle drug targeted delivery. The microcarriers carrying nanoparticle drugs would be targeting activated by local abnormal shear stress rate to release nanoparticle drugs. This study preliminarily demonstrates the feasibility of the shear-activated targeted nanoparticle drug delivery strategy in the treatment of aortic lesions, and the results may provide a theoretical basis for designing drug delivery systems and developing novel therapeutic methods.
