摘 要:目的:比较当归白凤颗粒减压水提与常压水提对各活性成分含量的影响,优选科学合理稳定可行的水提方法,为当归白凤颗粒工业化生产提供依据。方法:以当归白凤颗粒中活性成分阿魏酸、苦杏仁苷、橙皮苷、6-姜辣素以及干浸膏得率为评价指标,通过单因素考察确定水提次数和粉碎度,L_(9)(3~4)正交设计试验优化减压水提工艺参数:沸腾压力、沸腾温度、加水量、水提时间,减压水提优化后的3批验证试验与常压0.1 MPa、100℃水提工艺(浸泡时间、加水量、水提时间、次数、粉碎度皆同减压提取)比较各活性成分含量的差异。结果:减压水提最佳工艺参数为:水提压力降低到0.07 MPa最佳,水提温度65℃,第一次加水量为药材量的10倍,浸泡2.0 h,水提时间1.5 h,第二次加水量为药材量的8倍,水提时间1.0 h。减压水提各活性成分含量远高于常压水提(P<0.001),减压水提阿魏酸含量是常压水提的1.52倍,苦杏仁苷含量是其1.46倍,橙皮苷含量是其1.96倍,6-姜辣素含量是其1.35倍。结论:当归白凤颗粒药材减压水提优于常压水提,减压水提工艺科学、合理、稳定、可行,可为当归白凤颗粒水提取工业化生产提供依据。Objective:To compare the effects of reduced-pressure and atmospheric-pressure aqueous extraction of Danggui Baifeng granules on the content of each active ingredient,and to select a scientific,reasonable and stable method for the industrial production of Danggui Baifeng granules.Methods:The active ingredients ferulic acid,amygdalin,hesperidin,6-gingerin and dry infusion yield of Danggui Baifeng granules were evaluated by single-factor investigation to determine the number of extraction and comminution degree,and orthogonal design tests were conducted to optimize the parameters of reduced-pressure extraction process including boiling pressure,boiling temperature,water addition and extraction time.The differences in the content of each active ingredient were compared between the optimized batch of three validation experiments and the atmospheric pressure 0.1 MPa,100℃water extraction process(soaking time,water addition,water extraction time,number of times,and crushing degree were all the same as the reduced pressure extraction).Results:The best process parameters for the reduced pressure extraction were as follows:the best extraction pressure was reduced to 0.07 MPa,the extraction temperature was 65℃,for the first time,the ratio of liquid to solid was 10,the soaking time was 2.0 h,and the extraction time was 1.5 h.For the second time,the ratio of liquid to solid was 8,and the extraction time was 1.0 h.The content of each active ingredient of the reduced-pressure water extraction was much higher than that of the normal-pressure water extraction(P<0.001),and the content of ferulic acid was 1.52 times higher than that of the normal-pressure water extraction,that of amygdalin was 1.46 times higher,that of hesperidin was 1.96 times higher,and that of 6-gingerin was 1.35 times higher in the normal-pressure water extraction.Conclusion:The reduced-pressure aqueous extraction of Danggui Baifeng granules is better than the atmospheric pressure aqueous extraction,and the reduced-pressure aqueous extraction process is scie
