冷冻样品高压灭菌探究

摘　　要：目的 探索不需解冻、直接高压、彻底灭菌冷冻样品的程序。方法 通过分析自建程序134℃、120 min运行过程中冷冻样品包裹中心实测温度的变化,获得可行的灭菌程序。通过运行程序,并在冷冻样品上、中上、中、中下、下层放置生物指示物对灭菌效果进行验证。结果 在灭菌温度设定值为134℃的情况下,30 min仅能使冷冻样品的上、中上、下层灭菌合格,60 min能使冷冻样品的所有层皆灭菌合格。结论 提示134℃、60 min可用于-80℃以上冷冻样品的高压灭菌。Objective To determine a sterilizing procedure for frozen samples without thawing.Methods A feasibl esterilizing procedure was speculated by analyzing the change of measured temperature in the center of the frozen samples package during the running process of the self-built program(134℃,120 min).The same number of frozen samples were prepared, and biological in dicators were placed in upper, upper-middle, middle, lower-middle and lower layers.The speculated was run to verify the sterilization effect.Results When the sterilization temperature was set at 134℃,only upper, upper-middle and lower layers of the frozen samples were qualified for sterilization for 30 min, while all layers were qualified for sterilization for 60 min.Conclusion It is suggested that program(134℃,60 min) is suitable for steam sterilization of frozen samples above-80℃.
