摘 要:为研究滑液囊支原体(Mycoplasma synoviae,MS)感染对鸡卵泡颗粒细胞孕酮分泌的影响,试验以蛋鸡卵泡颗粒细胞为模型,进行MS感染,用ELISA孕酮检测试剂盒检测鸡卵泡颗粒细胞培养上清孕酮量的变化,用qRT-PCR方法和Western blot方法检测细胞孕酮分泌相关基因mRNA和蛋白表达水平的变化。结果显示:随着MS感染鸡卵泡颗粒细胞时间延长,细胞分泌孕酮的水平逐渐下降;调控孕酮分泌的关键基因(FSHR、STAR、3β-HSD)mRNA表达水平和蛋白表达随着MS感染鸡卵泡颗粒细胞时间的增长而降低。结果表明,MS感染会抑制鸡卵泡颗粒细胞孕酮的分泌水平,使孕酮表达相关基因mRNA和蛋白表达水平降低,初步解释了MS感染造成蛋鸡产蛋率下降的原因。In order to study the effect of Mycoplasma synoviae(MS) on progesterone secretion in laying hens follicular granulosa cells, the follicular granulosa cells of laying hen were used for cell model and infected with MS. The progesterone levels in supernatant were evaluated by ELISA, and the mRNA and protein levels of progesterone secretion-related genes were detected by qRT-PCR and Western blot, respectively. The results showed that the level of progesterone secreted by follicular granulosa cells decreased gradually with the extension of MS infection time. The mRNA expression of key genes regulating progesterone secretion, including FSHR, STAR and 3β-HSD showed decreasing with the time of MS infection at both m RNA and protein levels. The results indicated that MS infection could decrease the expression of progesterone-related genes and inhibit the secretion of progesterone in follicular granulosa cells of laying hen, which preliminarily clarified the reasons for the decline of laying rate caused by MS infection.
