摘  要:为研究不同浓度葡萄糖对蛋鸡卵泡膜细胞增殖、雌二醇水平及类固醇合成相关基因表达的影响,试验采集35周龄产蛋高峰期苏禽绿壳蛋鸡的小黄卵泡,通过分离、消化和过滤等得到原代卵泡膜细胞,待细胞长至培养皿70%~80%时,将含10%胎牛血清(FBS)培养基更换为含0.5%FBS培养基培养24 h后,随机分成6组,每组3个重复,对照组不添加葡萄糖,试验组分别添加0.5、1.5、3.0、4.5、9.0 mg/mL葡萄糖,继续培养48 h。结果显示:3.0 mg/mL葡萄糖处理组细胞活力显著高于对照组、0.5、4.5和9.0 mg/mL处理组(P<0.05),雌二醇含量显著高于对照组、0.5和9.0 mg/mL葡萄糖处理组(P<0.05),细胞类固醇合成急性调节蛋白(StAR)和细胞色素P450单加氧酶11A1(CYP11A1)的mRNA相对表达量显著低于对照组和9.0 mg/mL葡萄糖处理组(P<0.05);0.5 mg/mL葡萄糖处理组细胞StAR mRNA相对表达量显著低于对照组(P<0.05),CYP11A1相对表达量显著低于对照组、1.5和9.0 mg/mL组(P<0.05)。研究提示:3.0 mg/mL葡萄糖培养蛋鸡卵泡膜细胞效果最优,可提高细胞活力,增加培养液中雌二醇含量。This study aimed to investigate the effect of different concentrationon of glucose on proliferation, estradiol level and expression of steroid synthesis related genes in chicken theca cells harvested from follicles. Small yellow follicles of 35-weekage Suqin green eggshell layer during the peak egg production period were collected. The follicular membrane cells were obtained by separating, digesting, and filtrating. When the cells grew to about 70% to 80% of the culture dish, the theca cells cultured for24 h after replacing the medium containing 10% FBS with 0.5% FBS were randomly assigned to 6 groups. The theca cells were cultured with different concentrations of glucose with 0, 0.5, 1.5, 3.0, 4.5, and 9.0 mg/mL for 48 h. The results showed that the viability of chicken theca cells cultured with 3.0 mg/mL glucose was significantly higher than those cultured with 0, 0.5, 4.5,9.0 mg/mL glucose(P<0.05). The estradiol content of chicken theca cells cultured with 3.0 mg/mL glucose was significantly higher than that cultured with 0, 0.5 and 9.0 mg/mL glucose(P<0.05). The expression of StAR and CYP11A1 mRNA of chicken theca cells cultured with 3.0 mg/mL glucose was significantly lower than those cultured with 0 and 9.0 mg/mL glucose(P<0.05). The expression of StAR m RNA of chicken theca cells cultured with 0.5 mg/mL glucose was significantly lower than that cultured with 0mg/mL glucose, and the expression of CYP11A1 mRNA was significantly lower than that cultured with 0, 1.5, and 9.0 mg/mL glucose(P<0.05). These results indicated that 3.0 mg/mL glucose was the best in the culture of chicken theca cells, which improved the cell viability and estradiol content in culture.
