摘  要:【目的】比较HT29和HCT116两株细胞系体外培养细胞增殖和在裸鼠体内移植后的成瘤情况。【方法】检测结直肠癌细胞HT29和HCT116的细胞活力,观察克隆形成细胞的形态,检测2株细胞p53和p21蛋白的表达水平;将2株细胞移植于裸鼠右侧腹股沟皮下,观察移植瘤的生长情况,记录裸鼠的体质量及移植瘤的体积。【结果】与HT29细胞相比,HCT116细胞在体外的增长率极显著升高(P<0.01),p21蛋白正常表达,p53蛋白表达水平极显著降低(P<0.01);在阿霉素存在的情况下,HCT116细胞中的p21蛋白表达水平极显著升高(P<0.01)。14 d内HCT116细胞移植量对裸鼠体质量无显著影响(P>0.05);单只细胞接种量为4×10^(5)和2×10^(6)个时,HT29细胞肿瘤体积显著高于HCT116细胞(P<0.05)。【结论】HCT116细胞在体外的增殖速率显著高于HT29细胞,但是在体内HT29细胞比HCT116细胞更易成瘤,其分子基础可能与HT29细胞中p53基因突变导致的p53蛋白高表达及其下游p21蛋白低表达有关;高浓度的癌细胞移植有利于肿瘤形成。本研究为结直肠癌动物模型的建立及其进一步研究奠定了基础。[Purpose]To compare the proliferation of HT29 and HCT116 cell lines cultured in vitro and the tumorigenesis after transplantation in nude mice.[Methods]The viability of colorectal cancer cells HT29 and HCT116 were detected,observing the morphology of clonogenic experimental cells,and detecting the expression levels of p53 and p21 proteins in the two cell lines.The two cell lines were transplanted under the right groin of nude mice,observing the growth of the transplanted tumor,and recording the weight and volume of the transplanted tumor.[Results]Compare with HT29 cells,the proliferation rate of HCT116 cells in vitro was extremely significantly higher(P<0.01),the expression level of p21 protein was normal,but the expression level of p53 protein was extremely significantly lower(P<0.01);in the presence of doxorubicin,the expression level of p21 protein in HCT116 cell line was extremely significantly higher(P<0.01).The number of trans-planted HCT116 cells within 14 days had no significant effect on the body weight of nude mice(P>0.05).Under the amount of inoculation of per mice by 4×10^(5) and 2×10^(6),the tumor volume of nude mice transplanted with HT29 cells was significantly higher than that of HCT116 cells(P<0.05).[Conclusion]In vitro,the proliferation rate of HCT116 cells is significantly higher than that of HT29 cells.However,in vivo,HT29 cells transplanted into nude mice are more likely to form tumors than HCT116 cells.The molecular mechanism may be related to the high expression of p53 protein and the low expression of downstream p21 protein caused by p53 gene mutation in HT29 cells.High-er concentrations of cancer cells are more conducive to tumor formation.This study provides a theor-etical basis for the construction of colorectal cancer animal model and its further application.
