摘 要:【目的】研究酿酒酵母中PAD1基因的功能,为研究酿酒酵母芳香族化合物代谢过程和途径奠定基础。【方法】对酿酒酵母PAD1基因进行电子克隆,再采用生物信息学分析法对PAD1基因进行分析预测。【结果】酿酒酵母PAD1基因位于Ⅳ染色体1510902~1511630,全长729 bp,可编码242个氨基酸;系统进化树分析表明:该基因编码的蛋白质与肉桂酸脱羧酶(AAA20484.1)最为接近,同源性高达99.59%;蛋白质化学式为C1208H1950N328O336S9,分子量约为26733.31 ku,是稳定的疏水蛋白,在线粒体中发挥作用。二级结构预测结果未发现明显卷曲螺旋区域,有部分跨膜区存在,二级结构元件以α-螺旋和随机卷曲为主,延伸链和β-转角在蛋白质中分散存在。【结论】酿酒酵母PAD1基因是编码芳香族羧酸脱羧酶的基因,编码的蛋白质结构稳定,可能在线粒体中参与代谢,与酿酒酵母芳香族羧酸的代谢和其对芳香族羧酸的耐受性存在直接联系。[Purpose]To study the function of the Saccharomyces cerevisiae PAD1 gene,laying the foundation for studying the metabolic processes and pathways of aromatic compounds in S.cerevisiae.[Methods]PAD1 gene of S.cerevisiae was electronically cloned,and then the gene analyzed and predicted by bioinformatics analysis.[Results]The S.cerevisiae PAD1 gene was located on chromosomeⅣ1510902-1511630,with a total length of 729 bp,it could encode 242 amino acids.Phylogenetic tree analysis showed that the protein encoded by this gene was the most similar to phenylacrylic acid decarboxylase(AAA20484.1),and the homology was up to 99.59%.The chemical formula of the protein was C1208H1950N328O336S9,and the molecular weight was about 26733.31 ku.It was a stable hydrophobic protein and played a role in mitochondria.The secondary structure prediction results did not find any obvious coiled helix region,but some transmembrane regions existed,and the secondary structural components were mainlyα-helix and random coil,extended strand andβ-bend were scattered in the protein.[Conclusion]The S.cerevisiae PAD1 gene,which encodes an aromatic carboxylic acid decarboxylase,encodes a structurally stable protein,and it may be involved in metabolism in the mitochondria and it is directly linked to the metabolism of aromatic carboxylic acids and their tolerance to aromatic carboxylic acids in S.cerevisiae.
