摘  要:目的:建立金沙藤中异槲皮苷、紫云英苷、槲皮素、山奈素的定量分析方法。方法:选择反相高效液相色谱(reversed-phase high performance liquid chromatography,RP-HPLC)法,色谱柱为Agilent C_(18)柱,规格为150 mm×4.6 mm,5μm;甲醇(B)-0.3%甲酸(A)为流动相进行梯度洗脱;所用流速是1.0 mL·min^(-1),柱温30℃,检测波长是360 nm,进样量是10μL。结果:25 min内异槲皮苷、紫云英苷、槲皮素、山奈素分离度好,线性关系良好,异槲皮苷、紫云英苷、槲皮素、山奈素的线性范围分别是0~0.5290μg(n=6)、0~1.1728μg(n=6)、0~0.0810μg(n=6)、0~0.2568μg(n=6),异槲皮苷、紫云英苷、槲皮素、山奈素的平均加样回收率分别为98.7%(RSD=0.80%)、97.7%(RSD=0.40%)、94.9%(RSD=2.14%)、98.1%(RSD=2.38%)。结论:本实验选用RP-HPLC法定量分析金沙藤中异槲皮苷、紫云英苷、槲皮素、山奈素的含量,重复性好,比较稳定,操作方便,此法适用于测定金沙藤中异槲皮苷、紫云英苷、槲皮素、山奈素的含量。Objective:To establish a quantitative method for the determination of isoquercitrin,astragalin,quercetin and kaempferol in lygodium japonicum(Thunb.)Sw.Methods:RP-HPLC method was selected,and the chromatographic column was Agilent C_(18)with a specification of 150 mm×4.6 mm,5μm.Gradient elution was performed for the mobile phase with methanol(B)-0.3%formic acid(A).The choice of flow rate was 1.0 mL·min^(-1),column temperature was30℃,detection wavelength was 360 nm,the injection volume was 10μL.Results:Within 25 minutes,isoquercitrin,astragalin,quercetin and kaempferol were well separated with a good linear relationship.The linear ranges of isoquercitrin,astragalin,quercetin and kaempferol were 0~0.5290μg(n=6)、0~1.1728μg(n=6)、0~0.0810μg(n=6)、0~0.2568μg(n=6)and the average recoveries of isoquercitrin,astragalin,quercetin and kaempferol were 98.7%(RSD=0.80%)、97.7%(RSD=0.40%)、94.9%(RSD=2.14%)、98.1%(RSD=2.38%).Conclusion:RP-HPLC used in this study to quantitatively analyze the contents of isoquercitrin,astragalin,quercetin and kaempferol in lygodium japonicum(Thunb.)Sw.was with good repeatability,stability and convenient operation.It is suitable for the determination of isoquercitrin,astragalin,quercetin and kaempferol in lygodium japonicum(Thunb.)Sw.
