摘  要:BECN1基因是细胞自噬的启动基因之一。检测该基因在细毛羊不同发育时期皮肤毛囊上的表达规律,可为细毛羊皮肤毛囊发育是否存在细胞自噬提供参考。利用实时荧光定量、免疫组化及免疫印迹法对BECN1基因分别在细毛羊休止期、生长期、退行期皮肤毛囊上的定位和表达进行了研究。结果表明,BECN1基因在细毛羊各个时期皮肤毛囊的内根鞘、外根鞘以及毛母质中均有阳性表达,但以退行期最为显著;在细毛羊各个时期的毛囊中均检测到BECN1基因的转录与蛋白表达,均以退行期表达量最高,且显著高于休止期和生长期(P<0.05),而生长期与休止期间的表达量无明显差异(P>0.05)。上述结果间接表明,细毛羊的毛囊周期中均存在细胞自噬现象,并以退行期最为明显。BECN1 gene is one of the genes that initiate autophagy.Detecting the expression of this gene in the skin hair follicles of fine-wool sheep at different developmental stages can provide a reference for the existence of autophagy in the development of skin hair follicles of fine-wool sheep.Therefore,the localization and expression of BECN1 gene in the hair follicles of fine-wool sheep skin in telogen,anagen and catagen phases were studied by real-time fluorescence quantification,immunohistochemistry and immunoblotting.The results showed that the BECN1 gene was positively expressed in the inner root sheath,outer root sheath and hair matrix of the skin hair follicles of fine-wool sheep at all stages,but the most significant in the degenerative stage.The expression level of protein was the highest in the regression phase and was significantly higher than that in the resting and growth phases(P<0.05),but there was no significant difference in the BECN1 gene transcription and protein expression levels between the growth and resting periods(P>0.05).The above research results indirectly indicate that autophagy exists in the hair follicle cycle of fine wool sheep,and is the most obvious in the catagen phase.
