摘 要:建立了石竹中山奈酚三糖苷含量的测定方法并检测石竹不同品种、不同花发育时期以及不同部位山奈酚三糖苷的成分含量。使用Agilent ZORBAX SB-C_(18)(250 mm×4.6 mm,5μm)色谱柱,具体色谱条件:A通道流动相:1.5%磷酸、40%乙腈、50%冰乙酸,B通道流动相:1.5%磷酸水溶液,A通道从10%~40%进行梯度洗脱。检测波长为267 nm,柱温为30℃,流速为0.8 mL/min,进样量为10μL。研究结果表明石竹16个品种中均含有山奈酚三糖苷成分,但是不同品种中含量有所差异,以‘云粉晕’样品中山奈酚三糖苷成分的含量最高,‘狂欢’含量最低。花中的含量与开放时期有关,随着花逐渐开放山奈酚三糖苷的含量呈下降趋势。不同部位的含量存在显著差异,其中花瓣的含量最高,茎的含量最低。因此,可以依据不同品种活性成分含量的差异,培育并栽培推广专用型的石竹品种,也可以根据目标化合物选择合适的收获时间以及采收植物的不同部位,为进一步开发利用石竹植物资源提供实验基础和依据。A method was established to determine the content of kaempferidetriglycoside in Dianthus,and to detect the contents of kaempferidetriglycoside in different varieties,different flower development stages and different parts of Dianthus.Agilent ZORBAX SB-C_(18)(250 mm×4.6 mm,5μm)column was used.The specific chromatographic conditions were as follows:mobile phase in channel A:1.5%phosphoric acid,40%acetonitrile,50%glacial acetic acid,mobile phase in channel B:1.5%phosphoric acid is soluble in pure water,gradient elution from 10%to 40%in channel A.The detection wavelength was 267 nm,the column temperature was 30℃,the flow rate was 0.8 mL/min,and the injection volume was 10μL.The results show that kaempferidetriglycoside is found in 16 varieties of Dianthus,but the content of kaempferidetriglycoside is different among different varieties.The content of kaempferidetriglycoside in‘YunFenyun’sample is the highest,and that in‘Carnival’sample is the lowest.The content of kaempferidetriglycoside in flowers is related to the opening period.There are significant differences among different parts,among which the content of petal is the highest and that of stem is the lowest.Therefore,the varieties of Dianthus can be cultivated and popularized according to the content differences of active components in different varieties,and the appropriate harvest time and different parts of the harvest plant can also be selected according to the target compounds.The results provide experimental basis for further development and utilization of Dianthus.
