摘 要:目的:研究瞬时受体电位通道M8(TRPM8)在支气管哮喘患者气道来源的巨噬细胞中的表达及其临床意义。方法:收集支气管哮喘患者和志愿者的支气管肺泡灌洗液,分离其中巨噬细胞培养,通过RT⁃PCR和Western Blot技术检测TRPM8 mRNA和蛋白水平;通过ELISA技术检测细胞上清液中白细胞介素⁃10(IL⁃10)和肿瘤坏死因子⁃α(TNF⁃α)。结果:哮喘组患者支气管肺泡灌洗液中巨噬细胞TRPM8 mRNA和蛋白水平均明显高于对照组(P<0.01)。哮喘组IL⁃10水平明显高于对照组(P<0.01),然而两组TNF⁃α蛋白水平却差异无统计学意义(P>0.05)。通过Spearman相关分析,TRPM8蛋白与IL⁃10蛋白间存在相关性(r=0.969,P<0.01)。然而,TRPM8蛋白与CRP、IgE、FeNO、嗜酸粒细胞以及TNF⁃α蛋白浓度间均未发现明显相关性(P>0.05)。结论:哮喘患者气道巨噬细胞中TRPM8 mRNA和蛋白增多。其可能通过影响IL⁃10参与哮喘的发病机制。Objective:To determine the expression of transient receptor potential melastatin subtype 8(TRPM8)in macrophages from the airway of the asthmatic patients and to explore the clinical value.Methods:Macrophages were isolated from the samples of bronchoalveolar lavage fluid(BALF)which were collected from asthmatic patients and control group.TRPM8 were detected by qRT⁃PCR and Western Blot.Interleukin⁃10(IL⁃10)and tumor necrosis factor⁃α(TNF⁃α)in the supernatant were detected by ELISA.Results:The level of mRNA and protein of TRPM8 in the macrophages from the BALF of asthmatic group was significantly higher than those of control group(P<0.01).The concentration of IL⁃10 in the supernatant from the BALF of asthmatic group was significantly higher than that of control group(P<0.01),while no difference was found in TNF⁃α(P>0.05)between the two groups.Spearman correlation analysis showed that TRPM8 was positively related to IL⁃10(r=0.969,P<0.01),but no correlation was found between TRPM8 and either of CRP,IgE,FeNO,eosinophils and TNF⁃α(P>0.05).Conclusion:An increase of TRPM8 mRNA and protein was iden⁃tified in the airway macrophages from asthmatic patients.TRPM8 may probably affect asthma by regulating IL⁃10.
