摘 要:通过建立体外人肝微粒体孵育模型,采用超高效液相色谱串联Q Exactive质谱检测合成大麻素5F-EDMB-PICA母药及其代谢产物。结果表明:5F-EDMB-PICA通过羟基化、二羟基化、酯水解、脱氟羟基化、酯水解加羟基化和酯水解加脱氟羟基化等代谢途径,共产生7种代谢产物,其中羟基化反应代谢产物(M1-A)、酯水解反应代谢产物(M2)以及脱氟羟基化反应代谢产物(M3)为合适的潜在代谢标志物。解析了合成大麻素5F-EDMB-PICA的体外代谢途径及其代谢产物,为该合成大麻素的生物监测和代谢表征提供理论依据和技术支持。The human liver microsome incubation model was established,and the metabolic biotransformation of synthetic cannabinoids was analyzed using ultra-high performance liquid chromatography coupled to quadrupoleOrbitrap high-resolution mass spectrometry. The results showed that the metabolites of 5F-EDMB-PICA via hydroxylation,dihydroxylation,ester hydrolysis,defluorination with hydroxylation reaction,ester hydrolysis with hydroxylation reaction and ester hydrolysis with defluorination hydroxylation reaction were detected. The metabolomic profile of synthetic cannabinoids was analyzed,and the hydroxylation reaction metabolite(M1-A),the ester hydrolysis reaction metabolite(M2)and the defluorohydroxylation reaction metabolite(M3)were found to be potential metabolic products. The current study provides the theoretical basis and technical support for the biomonitoring and metabolic characterization of the synthetic cannabinoid 5F-EDMB-PICA.
