痒螨病患兔下丘脑室旁核组织结构及其内5-HT的表达量变化研究

摘  要:为了探究不同病变评分痒螨病患兔下丘脑室旁核(PVN)的组织结构及其内5-羟色胺(5-HT)的表达,采用HE染色及免疫组化(SABC)法观察患病组(双耳病变评分分别为1分、2分和3分)和健康组(0分)下丘脑PVN的形态结构变化及其内5-HT表达量的变化。结果表明:与健康组(0分)相比,患病组(1分、2分和3分组)下丘脑PVN大细胞的组织结构均有不同程度的病变,且病变程度随病变评分的升高而加剧。此外,兔下丘脑PVN内5-HT阳性细胞定位于神经元的细胞质;与健康组相比,随着痒螨病病变评分的增加,患病组下丘脑PVN大细胞内5-HT的表达量极显著增加(P<0.01);与2分组相比,3分组PVN内5-HT的表达量减少。以上结果表明,痒螨病可导致兔下丘脑PVN组织结构发生损伤,且可使其PVN内5-HT的表达量呈规律性增加,提示下丘脑PVN内5-HT可能参与了兔痒螨病的发生与发展。In order to explore the tissue structure of the paraventricular nucleus(PVN)and its expression of 5-HT in rabbits with different lesion scores of psoroptic mange,HE staining and SABC method were used to observe the changes of morphological structure and expression changes of 5-HT in hypothalamic PVN of rabbits with psoroptic mange in diseased groups(binary lesions scores of 1,2,and 3)and healthy group(score 0).The results showed that,compared with healthy group(score 0),the tissue structure of the hypothalamic PVN magnocellular and parvocellular cells in the diseased groups(score 1,2,and 3)all had different degrees of lesions,and the degree of lesions increased with the rising of lesion score.In addition,positive cells of 5-HT in rabbit hypothalamus PVN were positioned in the cytoplasm of neurons.Compared with healthy group(score 0),the expression quantity of 5-HT in the hypothalamic PVN magnocellular cells in the diseased group was extremely significantly increased(P<0.01)with the increase of psoroptic acariasis lesion score.Compared with score 2 group,the expression quantity of 5-HT in the PVN of score 3 group was reduced.The above results indicated that psoroptic mange could cause the related damage to the PVN tissue structure of the rabbit hypothalamus,and it could increase the expression of 5-HT in the PVN regularly,which suggested that 5-HT in the PVN of the hypothalamus might participate in the occurrence and development of rabbits psoroptic mange.
