摘　　要：为测定剑河白香猪IL-18核苷酸序列结构,采集剑河白香猪脾脏,提取总RNA,反转录成cDNA,通过PCR法扩增cDNA,将从凝胶中收集的DNA与PMD19-T Vector连接,转入E.coli DH5a感受态细胞,将感受态细胞在含有X-Gal、IPTG、Amp的L-琼脂平板培养基上培养,挑选单个白色菌落,用PCR法确认重组质粒中IL-18基因长度大小。结果表明:测得IL-18全基因长度为579 bp,编码192个氨基酸,与荣昌猪、内江猪、GenBank的序列AY262109、AY262109进行比较,核苷酸同源性分别是99.1%、99.5%、99.8%、99.8%,推导的氨基酸同源性分别是98.4%、99.5%、100%、100%。In order to determine the structure of the IL-18 nucleotide sequence in Jianhe Baixiang pig.The spleen of Jianhe Baixiang pig was collected,the total RNA was extracted,reverse transcribed into cDNA,amplified cDNA by PCR method,the DNA that collected from the gel connected with PMD19-T Vector,and it was transfered into E.coli DH5a competent cells,the competent cells were cultured on L-agar plate medium that containing X-Gal,IPTG,and Amp,a single white colony was selected,and the length of the IL-18 gene in the recombinant plasmid was confirmed by PCR.The whole length of IL-18 gene was 579 bp,encoding 192 amino acids,compared with Rongchang pig,Neijiang pig,AY262109 of GenBank and AY262109 of GenBank sequence,the nucleotide homology is 99.1%,99.5%,99.8% and 99.8%,the deduced amino acid homology is 98.4%,99.5%,100% and 100%.
