摘 要:目的探究地黄多糖(RGP)对慢性肾病(CKD)大鼠冠脉平滑肌细胞自噬活性、经典瞬时受体电位蛋白(TRPC)1/6表达的影响。方法45只大鼠随机分为对照组、CKD组和CKD+RGP组,每组各15只。利用5/6肾切除法诱导CKD大鼠冠脉钙化。RGP(100 mg/kg,8周)通过灌胃干预。检测各组血清中血肌酐、尿素氮、钙、磷水平,冠脉钙化程度,以及冠脉平滑肌细胞中TRPC1/6和自噬标志蛋白(LC3B和Beclin1)的表达水平。结果CKD组和CKD+RGP组大鼠血肌酐和尿素氮水平比较差异无统计学意义(P>0.05),且均显著高于对照组(P<0.05);3组大鼠血清中Ca2+和磷水平比较差异无统计学意义(P>0.05)。CKD组冠脉钙化水平和TRPC1/6蛋白表达水平显著高于对照组,LC3B和Beclin1蛋白水平显著低于对照组(P<0.05);CKD+RGP组冠脉钙化水平和TRPC1/6蛋白表达水平显著低于CKD组,LC3B和Beclin1蛋白水平显著高于CKD组(P<0.05);LC3B和Beclin1蛋白水平与TRPC1蛋白(分别为r=-0.532、r=-0.493)、TRPC6蛋白(分别为r=-0.601、r=-0.445)均呈负相关(P<0.05)。结论RGP缓解CKD大鼠冠脉钙化,抑制冠脉VMSCs中TRPC1/6蛋白的表达并促进自噬。Objective To explore the effects of Rehmannia glutinosa polysaccharide(RGP)on the autophagy and the expression of transient receptor potential channel(TRPC)1/6 levels of coronary smooth muscle cells in rats with chronic kidney disease(CKD). Methods The rats were randomly divided into control group,CKD group and CKD+RGP group(n=15). 5/6 nephrectomy was used to induce coronary artery calcification in CKD rats. RGP(100 mg/kg,8 weeks)was intervened by gavage. Serum creatinine,urea nitrogen,calcium and phosphorus levels,coronary calcification,and the expression levels of TRPC1/6 and autophagy marker proteins(LC3B and Beclin1)in coronary smooth muscle cells were detected.Results There was no significant difference in serum creatinine and urea nitrogen levels between the CKD group and CKD+RGP group(P>0.05),and they were significantly higher than those in the control group(P<0.05). There was no significant difference in serum calcium2+and phosphorus levels among the three groups of rats(P>0.05). The coronary calcification level and TRPC1/6 protein expression levels in the CKD group were significantly higher than those in the control group,and the protein levels of LC3B and Beclin1 were significantly lower than those in the control group(P<0.05). The coronary calcification level and TRPC1/6 protein expression levels in the CKD+RGP group were significantly lower than those in the CKD group,and the protein levels of LC3B and Beclin1 were significantly higher than those in the CKD group(P<0.05). LC3B and Beclin1 protein levels were negatively correlated with TRPC1 protein(r=-0.532,-0.493)and TRPC6 protein(r=-0.601,-0.445)(P<0.05). Conclusion RGP alleviates coronary calcification in CKD rats,inhibits the expression of TRPC1/6protein in coronary VMSCs and promotes autophagy.
