摘 要:目的探讨地黄叶水提物和地黄叶80%乙醇提取物(醇提物)的滋阴作用。方法小鼠按体质量被随机分为对照组、模型组、以及地黄叶水提物低剂量组(3 g/kg)、地黄叶水提物高剂量组(6 g/kg)、地黄叶醇提物低剂量组(3 g/kg)、地黄叶醇提物高剂量组(6 g/kg)、六味地黄丸组(0.6 g/kg)各给药组;每天灌服左旋甲状腺素钠持续14 d造小鼠阴虚模型,除对照组外,其余各组均采用同样的造模处理,各给药组在造模的当天灌服相应的受试药物,模型组和对照组灌服同等容积的溶媒蒸馏水;通过对小鼠大体观察、体质量、饮水量、肛温、肾脏指数等指标的检测分析评价地黄叶的水提物和醇提物的滋阴作用。结果造模7~14 d,小鼠出现毛发稀疏、枯槁、大便干燥等不同程度的阴虚症状,小鼠体质量分别显著降低13.7%和17.7%,饮水量分别升高48.1%和79.3%,在造模14 d,小鼠肛温和肾脏指数分别显著升高3.8%和10.9%;在给药7d和14 d,各受试药物(地黄叶水提物低剂量、地黄叶水提物高剂量、地黄叶醇提物低剂量、地黄叶醇提物高剂量、六味地黄丸)均不同程度缓解了造模诱导的阴虚症状,使小鼠体质量分别显著升高5.5%和5.4%、11.8%和13.8%、10.7%和12.1%、12.2%和13.8%、14.0%和19.5%,饮水量分别下降17.5%和32.7%、22.5%和36.5%、20.0%和30.8%、20.0%和34.6%、27.5%和40.4%,在给药14 d,小鼠肛温分别显著下降2.1%、2.9%、1.8%、2.9%和3.2%,而各受试药物对模型引起的肾脏指数升高均无显著影响。此外,地黄叶水提物高剂量组和地黄叶醇提物高剂量组各项指标比较总体无明显差异,与六味地黄丸组比较也总体相当。结论地黄叶水提物和醇提物均有滋阴作用,均以高剂量为佳,且在高剂量下,二者的作用相当,均与六味地黄丸的作用总体相当。To initially explore the nourishing-Yin effect of the water extract and 80% ethanol extract of Dihuang(Rehmannia glutinosa)leaves(RGL).Methods According to the body weight,mice were randomly divided into control group,model group and several administration groups including Liuwei Dihuang Pills(六味地黄丸)(0.6 g/kg),low dose of RGL water extract(3 g/kg),high dose of RGL water extract(6 g/kg),low dose of RGL ethanol extract(3 g/kg)and high dose of RGL ethanol extract(6 g/kg).Levothyroxine sodium was administered daily for 14 days to create a mouse model of Yin deficiency.Except for the control group,the other groups were treated with the same model.On the day of the model,the corresponding test drug was given,and the model group and the control group were given the same volume of solvent distilled water.The nourishing-Yin effect of the water and ethanol extracts of RGL was evaluated by the general observation of mice,body weight,water consumption,rectal temperature,kidney index and other indicators.Results After 7 or 14 days of modeling,the mice developed symptoms of Yin deficiency such as thinning of hair,hagginess and dry stool.The weight of the mice was significantly reduced by 13.7%and 17.7%,and the water consumption was increased by 48.1%and 79.3%,respectively.On the 14th day of modeling,the rectal temperature and kidney index of the mice were significantly increased by 3.8%and 10.9%,respectively.On the 7th and 14th day of administration,the test drugs(low dose of RGL water extract,high dose of RGL water extract,low dose of RGL ethanol extract,high dose of RGL ethanol extract,and Liuwei Dihuang Pills)all alleviated the symptoms of Yin deficiency induced by the modeling to varying degrees,and significantly increased the weights of the mice by 5.5%and 5.4%,11.8%and 13.8%,10.7%and 12.1%,12.2%and 13.8%,and 14.0%and 19.5%,and water consumption decreased by 17.5% and 32.7%,22.5% and 36.5%,20.0%and 30.8%,20.0%and 34.6%,27.5%and 40.4%,respectively.After 14 days of administra tion,the rectal temperature of mice de
