摘 要:目的研究紫苏梗乙酸乙酯提取物对D-半乳糖诱导小鼠衰老的干预作用。方法将48只小鼠随机分为正常对照组8只和造模组40只,采用颈背部皮下注射D-半乳糖构建小鼠衰老模型,然后将其分为模型组,阳性对照组和紫苏梗乙酸乙酯提取物低、中、高剂量组。阳性对照组灌胃200 mg/kg Vc,低、中、高剂量组分别灌胃200、400、800 mg/kg紫苏梗乙酸乙酯提取物溶液,连续给药4周。末次给药后,检测各组小鼠肝脏、心脏、脾脏指数,检测小鼠血清和肝脏、心脏、脾脏中丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽-过氧化物酶(GSH-Px)水平。结果紫苏梗乙酸乙酯提取物低、中、高剂量组小鼠肝脏、心脏、脾脏指数均高于衰老模型组,各给药组均能降低衰老小鼠血清和肝脏、心脏、脾脏中MDA含量,并能提高血清和3种脏器中SOD、GSH-Px活力,差异均具有统计学意义(P<0.01,P<0.05)。结论该结果说明紫苏梗乙酸乙酯提取物可通过降低体内脂质过氧化反应,调节MDA及SOD、GSH-Px水平达到抗衰老作用。Objective In order to study the effect of ethyl acetate extract from Zisugeng(Caulis Perillae)on the mice model of senescence induced by D-galactose.Methods A total of 48 mice were randomly divided into normal control group with 8 mice and model group with 40 mice.The mice model of senescence was established by subcutaneous injection of D-galactose solution at the neck and back,and then the model group was further randomly divided into the senescence model group,positive control group and ethyl acetate extract from Zisugeng(Caulis Perillae)low,middle and high dose groups,with 8 mice in each group.The positive control group was given 200 mg/kg Vc,and the ethyl acetate extract from Zisugeng(Caulis Perillae)low,middle and high dose groups were given the corresponding doses(200,400 and 800 mg/kg)via intragastric administration for 4 consecutive weeks.After the last administration of the medicines,the liver,heart and spleen indexes were calculated,and the levels of malondialdehyde(MDA),super-oxide dismutase(SOD)and glutathione peroxidas(GSH-Px)in serum,liver,heart and spleen tissue were measured.Results The indexes of liver,heart and spleen of ethyl acetate extract from Zisugeng(Caulis Perillae)groups were significantly higher(P<0.01,P<0.05)compared with those of the senescence model group.Meanwhile,the ethyl acetate extract from Zisugeng(Caulis Perillae)can significantly reduce the level of MDA,and increase the levels of SOD and GSH-Px in serum,liver,heart and spleen tissue(P<0.01,P<0.05).Conclusion Therefore,the ethyl acetate extract from Zisugeng(Caulis Perillae)exhibited the anti-senescence effect on the senescence mice induced by D-galactose by reducing the body’s lipid peroxidation response and regulating the levels of MDA,SOD and GSH-Px.
