摘  要:急性肝衰竭(acute liver failure,ALF)病情危重,病死率高,内科综合治疗疗效尚不满意,肝移植受限于肝源供给不足难以广泛开展。中药治疗可减轻肝脏损伤,延缓病情发展,降低病死率。基于动物模型积极筛选有效中药并阐述其作用机制具有重要意义。D-氨基半乳糖胺(D-galactosamine, D-GalN)可以引起肝细胞大量坏死,而脂多糖(lipopolysaccharide, LPS)可以模拟肝细胞大量死亡后的机体免疫应答,LPS联合D-GalN诱导建立的ALF模型更符合临床实际。因此,基于LPS/D-GalN构建的ALF模型,归纳总结可有效干预ALF的常见中药及其有效活性成分,并详细阐述其作用机制,以期对中药有效性评价、试验思路及临床合理应用提供参考。Acute liver failure(ALF) is characterized by critical condition and high mortality.The curative effect of comprehensive medical treatment is not satisfactory, and liver transplantation is difficult to be widely carried out due to insufficient sources of transplantable liver.The treatment of traditional Chinese medicine(TCM) can alleviate liver injury, delay the development of the disease and reduce the fatality rate.It is of great significance to actively screen the effective traditional Chinese drugs based on animal model and explain its mechanism of action.D-galactosamine(D-GalN) can cause massive necrosis of hepatocytes, and lipopolysaccharide(LPS) can simulate the immune response after massive death of hepatocytes.The model of ALF induced by LPS in combination with D-GalN is more in line with clinical practice.Therefore, based on the model of ALF constructed by LPS in combination with D-GalN,this paper summarized the common TCM and its active components which can effectively interfere with acute liver failure, and described its mechanism in detail, in order to provide reference for the evaluation of the effectiveness, experimental ideas and rational clinical application of TCM.
