疏肝利胆和胃方对反流性食管炎大鼠IL-8及VIPmRNA表达影响的研究

摘　　要：目的探讨疏肝利胆和胃方对肝胃不和型反流性食管炎大鼠IL-8的表达水平及相关受体的影响,阐明该方治疗肝胃不和型反流性食管炎的作用机理。方法对80只健康SD大鼠,采取SPSS 22.0软件随机平均分成4组,每组20只,分别为空白对照组、反流性食管炎模型对照组、疏肝利胆和胃方中药干预组和艾速平阳性药对照组,用药后观察各组大鼠的生存状态、体质量等。同时,于治疗的第14天、第28天对每组大鼠进行处死,解剖取食管样本,观察病理变化及黏膜愈合进程,观察食管黏膜上皮细胞间隙;采用ELISA法测IL-8的表达。结果与模型组大鼠相较,中药组、艾速平组大鼠食管黏膜组织愈合率明显高于模型组,食管黏膜上皮细胞间隙均低于模型组,均有统计学差异(P<0.05);用药组大鼠食管黏膜组织中N0S的表达水平明显高于模型对照组,差异有统计学意义(P<0.05);模型组大鼠食管黏膜组织中VIPmRNA表达水平与空白组比较,明显高于空白组,差异有统计学意义(P<0.05);中药组、西药组大鼠食管黏膜组织组织中IL-8表达水平与模型组比较,明显降低,差异有统计学意义(P<0.05)。结论疏肝利胆和胃方可能通过降低模型大鼠的IL-8相应受体的表达来治疗反流性食管炎。Objective To investigate the ffects of Shugan Lidan Hewei Reecipe(疏肝利胆和胃方)on the expression levels of IL-8 and related receptors in rats with disagreeable relux esophagitis of liver and stomach and clarify the mechanism of the treatment of disagreeable reflux esophagitis of liver and stomach.Methods Eighty healthy SD rats were randomly divided into 4 groups with 20 rats in each group using SPSS 22.0 software.The rats were divided into blank control group,reflux esophagitis model control group,Shugan Lidan Hewei Recipe intervention group,and Esomeprazole Magnesium Enteric-coated Capsules positive control group,respectively.The living status and weights of the rats in each group were observed after treatment.At the same time,the rats in each group were sacrificed on the 14th and 28th day of treatment,and the esophageal samples were dissec-ted to observe the pathological changes and mucosal healing process,and the esophageal mucosal epithelial cell space was ob-served.Westerm-blot was used to detect the expressions of its related rceplor proteins.The expression of IL.-8 was measured by ELISA.Results Compared with that of the model group,the healing rate of the esophageal mucosa tssues of the rats in the Shu-gan Lidan Hewei Recipe intervention group and the Esomeprazole Magnesium Enteric-coated Capsules positive control group was significantly higher and the cell gap of the esophageal mucosa epithelium was all lower than that in the model group,showing sta-tistical dfferences(P<0.05).The expression level of nitric oxide synthase(NOS)in the esophageal mucosal issues of rats in the treatment group was significantly higher than that in the model control group,showing statistical difference(P<0.05).The protein expression levels of its receptor VPAC1 in the esophageal mucosal tissues of rats in the Shugan Lidan Hewei Recipe inter-vention group and the Esomeprazole Magnesium Enteric-coated Capsules positive control group were significantly higher than that in the model group,showing statistical diference(P<0.
