泻白散化学成分的高效液相色谱-四级杆飞行时间串联质谱法分析

摘　　要：目的采用高效液相色谱-四级杆飞行时间串联质谱法(UPLC-Q-TOF-MS/MS)对经典名方泻白散中主要化学成分进行鉴定。方法色谱条件为ACQUITY UPLC BEH C1.7μm,2.1 mm×100 mm色谱柱,流动相0.1%甲酸水(A)-0.1%甲酸乙腈(B)梯度洗脱(0 min,5%B;3.5 min,15%B;6~6.5 min,30%B;12~12.5 min,70%B;18~25 min,100%B;26~30 min,5%B),进样体积为5μL,流速为400μL·min^(-1),柱温40℃。质谱条件选择电喷雾电离源(ESI),正、负离子2种检测模式,质量扫描范围m/z 100~1500。采用Progenesis QI软件、PubChem以及中国上海BioTree生物科技有限公司的BioTree中药数据库,并参考文献信息,进行泻白散中主要化学成分的鉴定。结果从泻白散中共鉴定出162个成分,主要包括有机酸类成分45个,黄酮类成分44个,生物碱类成分21个,酚酸类成分13个,糖苷类成分12个,香豆素类成分8个,内酯类成分6个,其他类成分11个,未知成分2个。结论通过UPLC-Q-TOF-MS/MS建立了一种可以快速、准确鉴别泻白散中化学成分的方法,为进一步研究泻白散的药效物质基础以及质量控制等方面提供理论依据。Objective To identify the main chemical constituents of Xiebai Powder(泻白散)by high performance liquid chromatography-quadrupole time of flight tandem mass spectrometry(UPLC-Q-TOF-MS/MS).Methods The chromatographic conditions were ACQUITY UPLC BEH Cig column(1.7μm,2.1 mm×100 mm),mobile phase 0.1%formic acid water(a)-0.1%formic acid acetonitrile(b)gradient elution(0 min,5%B;3.5 min,15%B;6~6.5 min,30%B;12~12.5 min,70%B;18~25 min,100%B;26~30 min,5%B),and the injection volume was 5μL.The flow rate was 400μL:min-and the column temperature was 40℃.Electrospray spray ionization source(ESI)and two detection modes of positive and negative ions were selected for mass spectrometry.The mass scanning range was m/z 100~1500.The main chemical components of Xiebai Powder were identified by using the Progenesis QI software,PubChem and BioTree Chinese medicine database of China Shanghai BioTree Biotechnology Co.,Ltd.,and the reference information.Results A total of 162 components were identified from Xiebai Powder,including 45 organic acids,44 flavonoids,21 alkaloids,13 phenolic acids,12 glycosides,8 coumarins,6 lactones,11 other components and 2 unknown components.Conclusion UPLC-Q-TOF-MS/MS is a rapid and accurate method for the identification of chemical constituents in Xiebai Powder,which provides a theoretical basis for further study on the pharmacodynamic material basis and quality control of Xiebai Powder.
