类叶升麻苷调控miR-496表达对高糖诱导小鼠肾脏足细胞损伤的影响

摘  要:目的研究类叶升麻苷对高糖诱导的小鼠肾脏足细胞(MPC5)损伤的影响,并探讨其潜在分子机制。方法采用不同浓度的类叶升麻苷作用于高糖诱导的MPC5细胞,流式细胞术检测细胞凋亡率,蛋白质印记检测(Western blot)检测肾病蛋白(nephrin)、足突蛋白(podocin)、B细胞淋巴瘤-2(Bcl-2)和Bcl相关X蛋白(Bax)蛋白的表达水平,实时荧光定量PCR(RT-qPCR)检测miR-496表达。转染miR-496模拟物至MPC5细胞,检测上调miR-496对高糖诱导的MPC5细胞损伤的影响。结果类叶升麻苷作用于高糖诱导的MPC5细胞后,细胞凋亡率、Bax蛋白表达显著降低,nephrin、podocin和Bcl-2蛋白表达显著升高,miR-496表达显著升高(P<0.05)。上调miR-496表达后,高糖诱导的MPC5细胞中nephrin、podocin和Bcl-2蛋白表达显著降低,细胞凋亡率、Bax蛋白表达显著升高(P<0.05)。下调miR-496表达能够降低类叶升麻苷对高糖诱导的MPC5细胞凋亡率、nephrin、podocin、Bcl-2和Bax蛋白表达的影响(P<0.05)。结论类叶升麻苷可降低高糖诱导的MPCS损伤,其机制与上调miR-496表达有关。Objective To investigate the effects of Acteoside on high glucose-induced renal podocyte(MPC5)injury in mice,and to explore its potential molecular mechanism.Methods The high glucose-induced MPC5 cells were treated by different concentrations of Acteosides,then flow cytometry was used to detect the cell apoptosis rate;Western Blot detected the expression levels of nephrin,podocin,B-cell lymphoma 2(Bcl-2)and BCl-associated X protein(Bax)proteins;Real-time fluorescence quantitative PCR(RT-qPCR)detected miR-496 expression.Moreover the miR-496 mimic was transfected into MPC5 cells,and the effects of up-regulating miR-496 on the high glucose-induced MPC5 cell injury were detected by the methods mentoned above.Results After n high glucose-induced MPC5 cells were treated by Acteoside,the cell apoptosis rate and Bax protein expression levels were significantly decreased,however,the expression levels of nephrin,podocin and Bcl-2 protein as well as miR-496 expression levels were significantly increased(P<0.05).After up-regulating the expression levels of miR-496,the expression of nephrin,podocin and Bcl-2 proteins in high glucose-induced MPC5 cells,the cell apoptosis rate and Bax protein expression levels were significantly increased(P<0.05).Down-regulating miR-496 could reduce the effects of Acteoside on the apoptosis rate in high glucose-induced MPC5 cell,and the expression levels of nephrin,podocin,Bcl-2 and Bax proteins(P<0.05).Conclusion Acteoside can relieve the high glucose-induced renal podocyte damage in mice,and its action mechanism is related to the up-regulation of miR-496 expression.
