摘 要:水貂出血性肺炎是严重危害水貂健康的一类传染病,其中最主要的病原为铜绿假单胞菌。本研究对近些年流行于河北、山东及辽宁等地的水貂出血性肺炎的疑似铜绿假单胞菌感染病料进行了病原分离、鉴定,以便充分了解该类疫病病原的流行情况,为防控提供可靠科学依据。通过常规细菌分离鉴定方法分离病原,以玻片凝集反应鉴定其血清型,对分离菌株分别进行动物回归试验、毒力试验、免疫原性试验及攻毒保护试验。结果获得15株铜绿假单胞菌,分别命名为PA1~PA15,其中14株为血清G型,占93.3%,1株为血清B型,占6.7%。通过毒力试验确定致病菌株,然后分别进行了本动物回归试验、免疫原性试验及攻毒保护试验。致病力最强的1株为血清G型(PA4株,暂命名为DL1007株),肌肉注射致死率可达100%(5/5),经灭活后采用一定剂量免疫水貂后并攻毒,可对致死剂量菌株产生全部保护(5/5存活)。此次疑似水貂出血性肺炎病料分离的病原为铜绿假单胞菌,毒力最强株(DL1007)的自制疫苗对水貂具有较强的免疫保护作用。该毒株可作为后续水貂铜绿假单胞菌、肺炎克雷伯菌与巴氏杆菌三联灭活疫苗研制的候选株。Mink hemorrhagic pneumonia is a kind of infectious disease seriously endangering mink health.In the past,the main pathogen was Pseudomonas aeruginosa.This study isolated and identified the pathogen of suspected Pseudomonas aeruginosa infection of mink hemorrhagic pneumonia prevalent in Hebei,Shandong and Liaoning in recent years,so as to fully understand the epidemic situation of this kind of disease pathogens and provide a reliable scientific basis for prevention and control.The pathogens were isolated by routine bacterial isolation and identification methods,and their serotypes were identified by agglutination reaction.The isolated strains were determined by the animal regression test,virulence test,immunogenicity test and challenge protection test.Results showed 15 strains of Pseudomonas aeruginosa were obtained,named PA1,PA2,...,and PA15 respectively.Among them,14 strains were serum type G,accounting for 93.3%,and 1strain was serum type B,accounting for 6.7%;The pathogenic strains were determined by the virulence test,and then the animal regression test,immunogenicity test and challenge protection test were carried out respectively.The most virulent strain was serotype g(PA4 strain,temporarily named Dl1007 strain).The lethal rate of intramuscular injection can reach 100%(5/5).After inactivation,a certain dose of immune mink can protect all lethal doses of virus attack(5/5 healthy).The pathogens of suspected mink disease materials are Pseudomonas aeruginosa.The most virulent strain(DL1007)has strong immune protection against mink.The strain can be used as a candidate for the development of triple inactivated vaccine of mink Pseudomonas aeruginosa,Klebsiella pneumoniae and Pasteurella.
