摘 要:通过建立降植烷诱导的系统性红斑狼疮(SLE)的小鼠模型,明确狼疮丸对于SLE小鼠模型的治疗作用。30只6~8周龄雌性小鼠BALB/c被随机分为3组,即对照组(首日单次腹腔注射生理盐水0.5 mL)、模型组(首日单次腹腔注射降植烷0.5 mL)和狼疮丸治疗组(首日腹腔注射降植烷0.5 mL,第2天起狼疮丸每日按剂量灌胃)。注射前及注射后每月采用尿蛋白试纸测定小鼠尿蛋白1次;注射后1、2、3、4、5、6个月ELISA法检测小鼠血清中抗双链DNA抗体(anti-dsDNA)、抗核抗体(anti-ANA)、肿瘤坏死因子(TNF-α)、白介素-6(IL-6);第6个月测定IgG蛋白;6个月处死动物后,观察肾脏HE染色及免疫荧光染色后的变化。与对照组相比,模型组在腹腔注射降植烷1个月后出现尿蛋白升高。4个月时,77.7%小鼠尿蛋白含量达到+++。狼疮丸治疗组可显著改善模型组小鼠尿蛋白含量(P<0.05);ds-DNA、ANA、TNF-α、IL-6、IgG浓度均显著低于模型组(P<0.05、P<0.01和P<0.001);与模型组相比,治疗组小鼠肾脏中的IgG、CD3表达显著减少(P<0.001)。狼疮丸能降低降植烷诱导的SLE样小鼠模型中尿蛋白、dsDNA、ANA、TNF-α、IL-6和IgG的浓度,并能减轻肾脏的损伤程度,对SLE小鼠具有较好的治疗作用。To establish a systemic lupus erythematosus(SLE)mouse model induced by pristane,and to study the therapeutic effect of lupus pills on the SLE mouse model.Thirty 6-8 weeks old female mice BALB/c were randomly divided into three groups,namely the control group(a single intraperitoneal injection of 0.5 mL of normal saline on the first day);the model group(a single intraperitoneal injection of0.5 mL of pristane on the first day);Lupus pill treatment group(intraperitoneal injection of 0.5 mL of pristane on the first day;Lupus pill by intragastric administration every day from the second day).Before injection and once a month after injection,the urine protein of mice was detected by urine protein test strip;ELISA method was used to detect anti-double-stranded DNA antibody(anti-dsDNA antibody)in mouse serum at 1,2,3,4,5,and 6 months after injection.anti-nuclear antibody(anti-ANA),tumor necrosis factor alpha(TNF-α),interleukin-6(IL-6);The IgG protein was measured at the 6th month;after the animals were sacrificed at 6 months,the changes of kidneys were observed under the microscope after HE staining and immunofluorescence staining.Compared with the control group,the model group had increased urinary protein 1 month after intraperitoneal injection of pristane.At 4 months,77.7%of mice had urine protein content of+++.The lupus pill treatment group could significantly improve the urine protein content of mice in the model group(P<0.05);the concentrations of ds-DNA,ANA,TNF-α,IL-6 and IgG were significantly lower than those in the model group(P<0.05,P<0.01,P<0.001);Compared with the model group,the expressions of Ig G and CD3 in the kidneys of the mice in the treatment group were significantly reduced(P<0.001).It concluded that Lupus pills can reduce the concentrations of urinary protein,ds-DNA,ANA,TNF-α,IL-6 and Ig G in the SLE-like mouse model induced by pristane,and can reduce the degree of kidney damage,It has a good therapeutic effect on SLE mice.
