摘  要:本文采用腹腔注射四氯化碳(CCl_(4))的方法建立大鼠肝损伤模型,研究杜仲对四氯化碳诱导的肝损伤大鼠AMPK/JNK信号通路的抑制作用及肝脏保护作用。结果表明,与肝损伤大鼠模型比较,杜仲低、中、高剂量组及水飞蓟素组大鼠血清ALT、AST、ALP、GGT、TNF-α、IL-6及IL-1水平,肝组织MDA水平及AMPK、JNK mRNA水平,肝组织AMPK、JNK蛋白表达水平及免疫组化表达评分均显著降低(P<0.05),肝组织SOD活性显著升高(P<0.05),大鼠肝组织病理学明显改善。杜仲能够明显降低肝损伤大鼠体内炎症反应及氧化损伤,修复大鼠肝功能及病理学变化,其机制可能与调节AMPK/JNK信号通路有关。To study the inhibitory effect of Eucommia ulmoides on AMPK/JNK signal pathway and liver protection in rats with carbon tetrachloride-induced liver injury.Rats were randomly divided into the control group,liver injury model group,low,middle and high dose Eucommia ulmoides groups and silymarin group.Compared with the model group,the levels of serum ALT,AST,ALP,GGT,TNF-α,IL-6and IL-1,the levels of MDA,AMPK and JNK mRNA in liver tissue,the protein level of AMPK and JNK in liver tissue and the score of immunohistochemical expression in liver tissue of Eucommia ulmoides group were significantly lower than those in model group(P<0.05),while the activity of SOD in liver tissue was significantly increased(P<0.05).Liver histopathology was significantly improved.Eucommia ulmoides can significantly reduce the level of serum inflammatory factors in rats with liver injury,alleviate the injury of oxidative stress in liver tissue,and improve liver function and liver histopathological changes in rats.The mechanism may be related to the regulation of AMPK/JNK signal pathway.
