摘 要:本研究探究姜黄提取物对肾癌细胞ACHN的炎症反应及其对Nude裸鼠体内成瘤的调控作用。体内试验以Nude裸鼠为载体,皮下接种4×10^(6)个人肾癌细胞株ACHN,4周后建立体内移植瘤模型,分别灌胃10 mg/kg、50 mg/kg及100 mg/kg的姜黄提取物进行治疗,记录小鼠体重及死亡率,通过酶联免疫吸附试验(ELISA)检测小鼠移植瘤内肿瘤坏死因子-α(TNF-α)及白细胞介素-6(IL-6)水平。体外试验以人肾癌细胞株ACHN为载体,通过MTT试验检测姜黄提取物对人肾癌细胞株ACHN细胞活力的影响,通过乳酸脱氢酶(LDH)标准试剂盒检测姜黄提取物对ACHN细胞LDH释放的影响;通过荧光定量PCR(real-time PCR)试验检测ACHN细胞内炎性因子肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)及转化生长因子-β(TGF-β)以及小鼠肿瘤组织和ACHN细胞内NF-κB通路相关基因的表达。结果显示,与Model组相比,姜黄提取物处理显著降低小鼠体内瘤重、死亡率及增加小鼠体重,且50 mg/kg的姜黄提取物治疗组有最好的治疗效果。与Control组相比,姜黄提取物呈浓度依赖性抑制ACHN细胞的细胞活力,增加细胞内LDH释放。此外姜黄提取物在体内外上调肿瘤组织中炎性因子的表达水平,诱导产生炎性损伤,且姜黄提取物在体内外激活了肿瘤组织中NF-κB通路基因的表达。姜黄提取物能诱导ACHN肾癌细胞的炎症反应,抑制Nude裸鼠的体内成瘤,并且其可能是通过调控机体内的NF-κB通路来实现的。姜黄提取物对肾细胞癌有一定的抑制作用,具有成为肾细胞癌治疗药物的潜力。The study to investigate the inflammatory response of turmeric extract on renal cancer cells ACHN and its regulatory effect on tumorigenesis in Nude nude mice.In vivo assay was performed using Nude mice as the vehicle,and 4×10^(6) individual kidney cancer cell line ACHN was inoculated subcutaneously,and an in vivo transplantation tumor model was established after four weeks.The treatment was carried out by gavage of 10 mg/kg,50 mg/kg and 100 mg/kg of turmeric extract,and the body weight and mortality of the mice were recorded.The effect of turmeric extract on the viability of human kidney cancer cell line ACHN was measured by MTT assay,and the effect of turmeric extract on the release of LDH from ACHN cells was measured by lactate dehydrogenase(LDH)standard kit;Detection of inflammatory factors tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),and interleukin-10(IL-10)in ACHN cells by real-time PCR assay and transforming growth factor-β(TGF-β)and the expression of NF-κB pathway-related genes in mouse tumor tissue and ACHN cells.Compared with the Model group,turmeric extract treatment significantly reduced tumour weight,mortality and increased body weight in mice,and the treatment group with 50 mg/kg of turmeric extract had the best therapeutic effect.Compared with the control group,turmeric extract showed concentration-dependent inhibition of cell viability and increased intracellular LDH release in ACHN cells.In addition,turmeric extract upregulated the expression level of inflammatory factors in tumour tissues in vitro and induced inflammatory damage.Moreover,turmeric extract activated the expression of NF-κB pathway genes in tumour tissues in vitro and in vivo.Conclusion Turmeric extract induced inflammatory responses in ACHN kidney cancer cells and inhibited tumorigenesis in Nude nude mice in vivo,and it may do so by regulating the NF-κB pathway in the organism.The turmeric extract showed some inhibitory effects on renal cell carcinoma and has the potential to be a therapeutic agent for renal cell ca
