摘  要:目的检测婴幼儿血管瘤组织中的早期生长反应因子1(Egr-1)表达情况,探讨Egr-1在血管瘤增殖过程中的意义。方法选取2019年1月至2021年2月广西壮族自治区民族医院的21例婴幼儿血管瘤患儿的标本组织,其中增殖期10例,消退期11例。应用免疫组织化学染色观察Egr-1在组织中的表达。提取组织总蛋白并应用Western blot分析Egr-1、血管内皮生长因子(VEGF)和基质金属蛋白酶-9(MMP-9)的表达水平。免疫荧光双标实验分别观察VEGF、MMP-9和Egr-1表达的定位。结果所有样本组织病理形态均符合婴幼儿血管瘤诊断。在免疫组织化学染色结果中,Egr-1在增殖期血管瘤中成强阳性表达,高于消退期,差异有统计学意义(P<0.01);Western blot结果提示,增殖期血管瘤Egr-1、VEGF和MMP-9的表达高于消退期,差异有统计学意义(P<0.01)。免疫荧光双标结果提示,增殖期血管瘤组织中Egr-1荧光表达强阳性的部位VEGF和MMP-9的阳性表达亦较高,在Egr-1表达阴性的部位VEGF和MMP-9的表达微弱。结论Egr-1的表达在婴幼儿血管瘤增殖期组织中表达升高,高表达的Egr-1可能通过调控MMP-9和VEGF的转录促进血管瘤内皮细胞的增殖和新生血管形成。Objective To detect the expression of early growth response factor 1(Egr-1)in infantile hemangioma tissues,and to explore the significance of Egr-1 in the proliferation of hemangioma.Methods From January 2019 to February 2021,a total of 21 tissue samples of children with infantile hemangioma were collected from Minzu Hospital of Guangxi Zhuang Autonomous Region,including 10 of proliferative phase and 11 of involuting phase.Immunohistochemical staining was used to observe the expression of Egr-1 in tissues.Total protein was extracted and the expression levels of Egr-1,vascular endothelial growth factor(VEGF)and matrix metalloproteinase-9(MMP-9)were analyzed by Western blot.The expression and localization of VEGF,MMP-9 and Egr-1 were observed by double immunofluorescence assay.Results The morphology of all sample tissues were consistent with infantile hemangioma.The expression of Egr-1 in proliferative hemangioma was higher than that in regression hemangioma,and the difference was statistically significant(P<0.01).The results of Western blot showed that the expressions of Egr-1,VEGF and MMP-9 in the proliferative phase were higher than those in the regression phase,and the differences were statistically significant(P<0.01).The double immunofluorescence assay results demonstrated that in proliferative hemangioma,the VEGF and MMP-9 were upregulated when Egr-1 were present,while negative expression of Egr-1 has a lower VEGF and MMP-9 expression.Conclusion The expression of Egr-1 is increased in the tissues of infantile hemangioma during the proliferative stage,and the high expression of Egr-1 may promote the proliferation and neovascularization of hemangioma by regulating the transcription of MMP-9 and VEGF.
