摘 要:目的:研究治伤风颗粒对干酵母致热模型大鼠的退热作用及其机制。方法:将健康SD雄性大鼠随机分为正常组、模型组、布洛芬组和治伤风颗粒组,采取预防性给药方式,连续灌胃3 d。禁食不禁水6 h后测得基础体温,再灌胃1次,0.5 h后造模,每间隔1 h测肛温并记录,造模后5 h测肛温后再灌胃1次,末次测肛温后麻醉大鼠并收集其相关血清及下丘脑组织。采用酶联免疫吸附试验和生化法检测下丘脑和血清中白细胞介素(IL)1β、肿瘤坏死因子α(TNF-α)、IL-6、前列腺素E2(PGE2)、环磷酸腺苷(cAMP)、促肾上腺皮质激素释放激素(CRH)、精氨酸血管升压素(AVP)、去甲肾上腺素(NE)及α-黑素细胞刺激素(α-MSH)水平,采用苏木精-伊红染色观察下丘脑组织病理变化。结果:相比于模型组,治伤风颗粒组大鼠下丘脑和血清中IL-1β、TNF-α、IL-6、PGE2和cAMP含量,下丘脑CRH含量均明显降低(P<0.01);下丘脑和血清中AVP含量,下丘脑NE、α-MSH含量均明显升高(P<0.01),差异均有统计学意义。治伤风颗粒组大鼠神经元空泡变性、核固缩程度浅,仅有少量炎症细胞浸润。结论:治伤风颗粒对酵母致热模型大鼠有良好的解热作用,其机制与下调下丘脑和血清中的IL-1β、IL-6、TNF-α、PGE2和cAMP以及下丘脑中的CRH表达,上调下丘脑和血清中的AVP以及下丘脑中的NE和α-MSH的表达有关。OBJECTIVE: To study the antipyretic effects and mechanism of Zhishangfeng granules on dry yeast-induced fever model rats. METHODS: Healthy SD male rats were randomly divided into the normal group, model group, ibuprofen group and Zhishangfeng granules group, preventive administration was given for 3 d. The basal body temperature was measured after 6 h of fasting and water intake, and then gavage was administered once again. After 0.5 h, the model was established, and the rectal temperature was measured and recorded every 1 h. Anal temperature was measured 5 h after modeling and then administered. After the last rectal temperature measurement, the rats were anesthetized and the relevant serum and hypothalamus tissues were collected. The levels of interleukin(IL) 1β, tumor necrosis factor-α(TNF-α), IL-6, prostaglandin E2(PGE2), cyclic adenosine monophosphate(cAMP), corticotropin-releasing hormone(CRH), AVP, norepinephrine(NE) and α-melanocyte-stimulating hormone(α-MSH) levels in hypothalamus and serum were detected by enzyme-linked immunosorbent assay and biochemical methods. Hematoxylin-eosin staining was used to observe the pathological changes of hypothalamus. RESULTS: Compared with the model group, the levels of IL-1β, TNF-α, IL-6, PGE2, cAMP in hypothalamus and serum, and CRH in hypothalamus in the Zhishangfeng granule group decreased significantly(P<0.01);the levels of AVP in hypothalamus and serum, and the levels of NE and α-MSH in hypothalamus increased significantly(P<0.01), with statistically significant differences. The degree of neuronal vacuolar degeneration and karyopyknosis in the Zhishangfeng granules group was mild, and only a small amount of inflammatory cell infiltration was observed. CONCLUSIONS: Zhidongfeng granules have better antipyretic effects on yeast-induced fever model rats, and its mechanism is related to down-regulating the expression of IL-1β, IL-6, TNF-α, PGE2 and cAMP in hypothalamus and serum and CRH in hypothalamus, up-regulating the expression of AVP in hypothalamus and
