摘 要:采用硅胶柱色谱、反相C色谱、Sephadex LH-20凝胶柱色谱、高效液相色谱及中压半制备液相色谱等色谱分离技术对纤枝金丝桃中的化学成分进行分离纯化,通过MS、NMR等波谱数据结合理化性质鉴定化合物结构。从纤枝金丝桃乙酸乙酯部位中分离得到10个化合物,分别鉴定为lagarxanthone A(1)、1,7-二羟基(口山)酮(2)、3,4,5-三羟基(口山)酮(3)、2,7-二羟基-1-甲氧基(口山)酮(4)、1,3-二羟基-7-甲氧基(口山)酮(5)、1,5-二羟基-8-甲氧基(口山)酮(6)、3,4,-二羟基-2-甲氧基(口山)酮(7)、3,4-二羟基-5-甲氧基(口山)酮(8)、2,3-二甲氧基(口山)酮(9)、2,3,4-三甲氧基(口山)酮(10)。其中化合物1是未见报道的新化合物,化合物2~10是首次从该植物中分离得到。对10个化合物进行基于L6骨骼细胞的促葡萄糖摄取实验,结果显示所有化合物均显示弱的促葡萄糖摄取活性。Repeated silica gel column chromatography,reversed-phase Ccolumn chromatography,Sephadex LH-20 column chromatography,high performance liquid chromatography and semi-preparative medium pressure liquid chromatography were performed to separate and purify the chemical constituents of Hypericum lagarocladum.Spectroscopic methods such as mass spectrometry(MS)and nuclear magnetic resonance(NMR)combined with physicochemical properties were adopted in identifying the structure of the isolated compounds.Ten compounds were isolated from the ethyl acetate fraction of H.lagarocladum and identified as lagarxanthone A(1),1,7-dihydroxyxanthone(2),3,4,5-trihydroxyxanthone(3),2,7-dihydroxy-1-methoxyxanthone(4),1,3-dihydroxy-7-methoxyxanthone(5),1,5-dihydroxy-8-methoxyxanthone(6),3,4-dihydroxy-2-methoxyxanthone(7),3,4-dihydroxy-5-methoxyxanthone(8),2,3-dimethoxyxanthone(9),and 2,3,4-trimethoxyxanthone(10).Among them,compound 1 was a new compound,and compounds 2-10 were isolated from this plant for the first time.These ten compounds were tested for glucose uptake in L6 cells,and the results showed that all the compounds had no significant effect on glucose uptake.
