摘 要:为了探讨卵巢激素是否影响甘氨酰-tRNA合成酶(Gars)在小鼠子宫的表达,试验将12只小鼠切除卵巢后随机分成4组,分别为E2组(注射E2100 ng)、P4组(注射P42 mg)、E2+P4组(注射E2100 ng和P42 mg)和Oil组(注射芝麻油0.1 mL),采用mRNA原位杂交、实时荧光信号颜色定量PCR、免疫组织化学和Western-blot分析等方法,从基因和蛋白质水平对Gars在小鼠子宫的表达进行定位和定量研究。结果表明:加入碱性磷酸酶显色底物NBT/BCIP后,E2组信号颜色为深棕色(偏黑),P4组及E2+P4组为浅棕色,Oil组仍为绿色;与注射芝麻油相比,E2及E2+P4可上调切除卵巢小鼠子宫Gars基因的表达(P<0.05),P4不影响切除卵巢小鼠子宫Gars基因的表达(P>0.05);加入免疫组织化学显色底物DAB后,E2组、E2+P4组信号颜色为深棕色,Oil组为浅棕色,P4组为蓝色;与注射芝麻油相比,E2及E2+P4可上调切除卵巢小鼠子宫Gars蛋白的表达P<0.05),P4则显著下调切除卵巢小鼠子宫Gars蛋白的表达(P<0.05)。说明Gars的表达受到卵巢类固醇激素的调控。In order to investigate whether ovarian hormones affect the expression of glycyl-tRNA synthase(Gars) in murine uterus, twelve mice were randomly divided into 4 groups: E2 group(100 ng E2), P4 group(2 mg P4), E2+P4 group(100 ng E2 and 2 mg P4) and Oil group(0.1 mL sesame Oil). The expression of Gars in mouse uterus was mapped and quantified at gene and protein levels by in situ hybridization of mRNA, real-time fluorescence quantitative PCR, immunohistochemistry and Western-blot. The results showed that the E2 group was dark brown(slightly black), the P4 group and E2+P4 group were light brown, and the Oil group was still green after adding NBT/BCIP. Compared with Oil group, E2 group and E2+P4 group up-regulated the expression of Gars gene in the uterus of ovariectomized mice(P<0.05). There was no significant difference between E2 group and E2 + P4 group(P>0.05). There was no significant difference between P4 group and oil group(P>0.05). After adding immunohistochemical chromogenic substrate DAB, E2 group and E2 + P4 group showed dark brown, oil group was light brown and P4 group was blue. Compared with Oil group, E2 group and E2+P4 group up-regulated the expression of Gars protein in the uterus of ovariectomized mice(P<0.05), and P4 group significantly down-regulated the expression of Gars protein in the uterus of ovariectomized mice(P<0.05). It indicated that the expression of Gars was regulated by ovarian steroid hormones.
