摘 要:为了明确Notch家族及其配体在绵羊卵巢黄体组织和颗粒细胞中的表达及对激素合成的影响,试验采用实时荧光定量PCR、免疫荧光染色等方法,对绵羊卵巢不同时期黄体组织及颗粒细胞中Notch家族及其配体的表达差异,以及在添加Notch抑制剂后类固醇激素的变化进行了研究。结果表明:在体外成功培养了绵羊卵巢颗粒细胞。Notch2基因在颗粒细胞中相对表达量显著或极显著高于不同时期的黄体(P<0.05或P<0.01),而Notch1和Notch3基因在不同时期黄体中的相对表达量显著或极显著高于颗粒细胞(P<0.05或P<0.01);配体中Jag1基因在中期黄体及晚期黄体中的相对表达量极显著高于颗粒细胞(P<0.01),Jag2、Dll1基因在各时期黄体中的相对表达量均显著或极显著高于颗粒细胞(P<0.05或P<0.01)。在添加Notch抑制剂后,孕酮含量显著上调(P<0.05),而雌二醇含量没有明显变化(P>0.05)。说明Notch不同受体及其配体在绵羊卵巢黄体组织和颗粒细胞中的表达不尽相同,并且Notch信号通路对孕酮的合成发挥负调控作用。In order to clarify the expression of Notch family and its ligands in different types of cells of sheep ovary and their effects on hormone synthesis, the expression of Notch family and its ligands in luteal tissues and granulosa cells at different stages of sheep ovary were detected by real-time fluorescence quantitative PCR;immunofluorescence technic and the changes of steroid hormones after the addition of Notch inhibitors were detected by radioimmunoassay. The results showed that sheep granulosa cells were successfully cultured in vitro. The relative expression of Notch2 gene in granulosa cells was significantly or extremely significantly higher than that in corpus luteum at different stages(P<0.05 or P<0.01), while the relative expression of Notch1 and Notch3 gene in corpus luteum at different stages was significantly or extremely significantly higher than that in granulosa cells(P<0.05 or P<0.01). The relative expression levels of Jag1 gene in middle luteum and late luteum were extremely significantly higher than those in granule cells(P<0.01). The expression levels of Jag2 and Dll1 gene in each phase of luteum were significantly or extremely significantly higher than those in granule cells(P<0.05 or P<0.01). After Notch inhibitor was added, progesterone content was significantly up-regulated(P<0.05), while estradiol content had no significant change(P>0.05). These results indicated that the expressions of Notch receptors and their ligands were different in luteal tissues and granulose cells, and Notch signaling pathway played a negative regulatory role in the synthesis of progesterone.
