摘 要:目的探讨miRNA对激素敏感型特发性肾病综合征(INS)患儿调节性T细胞(Treg)的影响。方法选取40例激素敏感型INS患儿,根据病情分为INS初发组和INS缓解组,各20例;另选取20例同年龄健康体检儿童作为对照组。采用流式细胞术(FCM)检测三组外周血中辅助型T细胞2(Th2)、Treg细胞百分比及Treg分化相关miRNA表达水平并进行比较,分析外周血Treg、Th2细胞百分比的相关性。结果INS初发组患儿外周血Treg细胞百分比(3.45±1.88)%低于对照组的(5.99±2.01)%及INS缓解组的(5.62±1.44)%,差异均有统计学意义(P<0.05);INS初发组患儿外周血Th2细胞百分比(4.58±1.35)%高于对照组的(3.43±1.03)%及INS缓解组的(3.69±1.24)%,差异均有统计学意义(P<0.05);INS初发组患儿miRNA155基因表达(27.70±12.30)×10^(-5)低于对照组的(51.62±18.70)×10^(-5)及INS缓解组的(36.36±16.10)×10^(-5),差异均有统计学意义(P<0.05);INS缓解组与对照组miRNA155基因表达比较,差异无统计学意义(P>0.05);INS初发组患儿miRNA21基因表达(15.88±3.58)×10^(-5)低于对照组的(26.04±4.54)×10^(-5)及INS缓解组的(23.09±4.29)×10^(-5),差异有统计学意义(P<0.05);INS缓解组与对照组miRNA21基因表达比较,差异无统计学意义(P>0.05);INS初发组患儿miRNA31基因表达(20.63±6.45)×10^(-5)高于对照组的(13.37±4.38)×10^(-5)及INS缓解组的(15.47±3.29)×10^(-5),差异有统计学意义(P<0.05);INS缓解组与对照组miRNA31基因表达比较,差异无统计学意义(P>0.05)。相关性分析结果显示,外周血Treg百分比与Th2细胞百分比呈负相关关系(r=-0.527,P<0.05)。结论miRNA可能在激素敏感型INS患儿Treg比例变化中发挥着重要作用。Objective To discuss the effect of miRNAs on regulatory T cells(Treg)in children with hormone-sensitive idiopathic nephrotic syndrome(INS).Methods A total of 40 children with hormone-sensitive INS were divided into INS initial group and INS remission group according to their condition,with 20 cases in each group;another 20 healthy children of the same age were selected as the control group.Flow cytometry(FCM)was used to detect T helper 2(Th2),percentage of Treg cells and the expression levels of Treg differentiationrelated miRNAs in the peripheral blood of the three groups and compared them,and the correlation between the percentages of peripheral blood Treg and Th2 cells was analyzed.Results The percentage of Treg cells in the peripheral blood of children in the INS initial group was(3.45±1.88)%,which was lower than(5.99±2.01)%in the control group and(5.62±1.44)%in the INS remission group,and the difference was statistically significant(P<0.05).The percentage of Th2 cells in the peripheral blood of children in the INS initial group was(4.58±1.35)%,which was higher than(3.43±1.03)%in the control group and(3.69±1.24)%in the INS remission group,and the differences were statistically significant(P<0.05).The miRNA155 gene expression in the INS initial group was(27.70±12.30)×10^(-5),which was lower than(51.62±18.70)×10^(-5) in the control group and(36.36±16.10)×10^(-5) in the INS remission group,and the differences were all statistically significant(P<0.05).There was no statistically significant difference in miRNA155 gene expression between the INS remission group and the control group(P>0.05).The miRNA 21 gene expression of children in INS initial group was(15.88±3.58)×10^(-5),which was lower than(26.04±4.54)×10^(-5) in the control group and(23.09±4.29)×10^(-5) in the INS remission group,and the differences were all statistically significant(P<0.05).There was no statistically significant difference in miRNA 21 gene expression between the INS remission group and the control group(P>0.05).The miRNA 3
